- Description: The non-mevalonate isoprenoid pathway, also known as the 1-deoxy-D-xylulose-5- phosphate DXP pathway, is absent in humans, but present in the anopheles mosquito responsible for the transmission of malaria. DXP reductoisomerase - a key enzyme in the DXP pathway in Plasmodium falciparum (PfDXR) has been identified as a target for the design of novel anti-malarial drugs. Fosmidomycin and its acetyl analogue (FR900098) are known to be inhibitors of PfDXR and, in this study, synthetic variations of the fosmidomycin scaffold have led to four series of novel analogues. Particular attention has been centred on the introduction of various substituted benzyl groups in each of these series in order to occupy a recently discovered vacant pocket in the PfDXR active-site and thus enhance ligand-enzyme binding. In the process 160 ligands and precursors have been prepared, no less than 119 of them novel. Fistly, a series of C-benzylated phosphonate esters and phosphonic acids were synthesised, in which the fosmidomycin hydroxamate Mg2+- coordinating moiety was replaced by an amide funtionality and the number of methylene groups in the “hydrophobic patch” between the phosphonate and the hydroxamate moiety was decreased from two to one. Several approaches were explored for this series, the most successful involving reaction of 3- substituted anilines with a-bromo propanoic acid in the presence of the coupling agent 1,1'- carbonyldiimidazole (CDI), followed by Michaelis-Arbuzov phosphonation using triethyl phosphite. Reaction of the resulting chiral phosphonate esters with bromotrimethylsilane gave the corresponding phosphonic acids in good yields. In order to obviate chirality issues, a second series of potential “reverse” fosmidomycin analogues was synthesised by replacing the methylene group adjacent to the the phosphonate moiety with a nitrogen atom. Deprotonation, alkylation and phosphorylation of various amines gave diethyl #-benzylphosphoramidate ester intermediate. Aza-Michael addition of these intermediates, followed by hydrolysis gave the corresponding carboxylic acids which could be reacted with different hydroxylamine hydrochloride derivatives to afford the novel hydroxamic acid derivatives in good yields. Thirdly, a series of a novel #-benzylated phosphoramidate derivatives were prepared as aza- FR900098 analogues. Alkylation of different amines using bromoacetalde-hyde diethylacetal gave a series of N-benzyl-2,2-diethoxyethylamine compounds, which were then elaborated via a futher six steps to afford novel #-benzylated phosphoramidate derivatives. Finally, in order to ensure syn-orientation of the donor atoms in the Mg - coordinating group and, at the same time, introduce conformational constraints in the ligand, the hydrophobic patch and the hydroxamate moiety were replaced by cyclic systems. Several approaches towards the synthesis of such conformationally constrained phosphoramidate analogues from maleic anhydride led to the unexpected isolation of an unprecedented acyclic furfuryl compound, and 1H NMR and DFT-level theoretical studies have been initiated to explore the reaction sequence. A series of #-benzylated phosphoramidate derivatives containing dihydroxy aromatic rings (as the conformationally constrained groups) to replace the hydroxamate moiety, were successfully obtained in six steps from the starting material, 3,4-dihydroxylbenzaldehyde. While in vitro assays have been conducted on all of the synthesised compounds, and some of the ligands show promising anti-malarial inhibitory activity - most especially the conformationally constrained cyclic #-benzylated phosphoramidate series. Interestingly, a number of these compounds has also shown activity against T.brucei - the causative agent of sleeping sickness. In silico docking studies of selected compounds has revealed the capacity of some of the ligands to bind effectively in the PfDXR active-site with the newly introduced benzyl group occupying the adjacent vacant pocket. The physico-chemical properties of these ligands were also explored in order to predict the oral-bioavailability. - Description: Considerable efforts have been made in the modification of existing antimalarial drugs, and the support of incentive programmes have led to a drastic decrease in malaria cases reported by WHO during the past 6 years. However, the development of drug resistance threatens the eradication of this deadly disease and has prompted research on the synthesis of novel antimalarial drugs. Our research has involved the design and synthesis of novel benzylated phosphonate esters as potential 1-deoxy-D-xylose-5-phosphate reductoisomerase (DXR) inhibitors. A series of amidoalkylphosphonate esters were obtained by reacting various 3-subsituted anilines and heterocyclic amines with chloroalkanoyl chlorides and reacting the resulting chloroalkanamides with triethyl phosphite using Michaelis-Arbuzov methodology. Benzylation of the phosphonate esters afforded a series of novel N-benzylated derivatives in good yields and these compounds were fully characterised by NMR and HRMS methods. Several approaches to the introduction of a benzyl group at the C-2 position of the phosphonate ester derivatives have been explored, leading unexpectedly to the isolation of unprecedented tetrahydrofuranyl derivatives. Studies towards the preparation of potential bi-functional PfDXR / HIV-1 RT inhibitors have also been initiated. Preliminary in silico docking studies of selected non-benzylated and benzylated phosphonated derivatives into the Pf-DXR active-site has provided useful insight into the binding potential of these ligands. Bioassays have revealed a very low toxicity for all the synthesised phosphonated compounds and a number of these ligands also exhibit a promising inhibitory activity against the Plasmodium parasite.

- Description: HSP70 is one of the most important families of molecular chaperone that regulate the folding and transport of client proteins in an ATP dependent manner. The ATPase activity of HSP70 is stimulated through an interaction with its family of HSP40 co-chaperones. There is evidence to suggest that specific partnerships occur between the different HSP40 and HSP70 isoforms. While some of the residues involved in the interaction are known, many of the residues governing the specificity of HSP40-HSP70 partnerships are not precisely defined. It is not currently possible to predict which HSP40 and HSP70 isoforms will interact. We attempted to use bioinformatics to identify residues involved in the specificity of the interaction between the J domain from HSP40 and the ATPase domain from the HSP70 isoforms from humans. A total of 49 HSP40 and 13 HSP70 sequences from humans were retrieved and used for subsequent analyses. The HSP40 J domains and HSP70 ATPase domains were extracted using python scripts and classified according to the subcellular localization of the proteins using localization prediction programs. Motif analysis was carried out using the full length HSP40 proteins and Multiple Sequence Alignment (MSA) was performed to identify conserved residues that may contribute to the J domain – ATPase domain interactions. Phylogenetic inference of the proteins was also performed in order to study their evolutionary relationship. Homology models of the J domains and ATPase domains were generated. The corresponding models were docked using HADDOCK server in order to analyze possible putative interactions between the partner proteins using the Protein Interactions Calculator (PIC). The level of residue conservation was found to be higher in Type I and II HSP40 than in Type III J proteins. While highly conserved residues on helixes II and III could play critical roles in J domain interactions with corresponding HSP70s, conserved residues on helixes I and IV seemed to be significant in keeping the J domain in its right orientation for functional interactions with HSP70s. Our results also showed that helixes II and III formed the interaction interface for binding to HSP70 ATPase domain as well as the linker residues. Finally, data based docking procedures, such as applied in this study, could be an effective method to investigate protein-protein interactions complex of biomolecules.

The garden as a metaphor for paradise

- Description: In this half thesis the use of the garden as a metaphor for paradise has been explored. The English word “ paradise“ was derived from the Greek word “ paradeisos” which in turn was derived from the Old Avestana “ pairi-daeza,” meaning an enclosure. In Ancient Persia the concept applied to an enclosed garden in the modern sense of the word. For this reason the thesis begins with an examination of the development of the garden in this desert region. A more-or-less continuous chain of development in both the physical and allegorical nature of the garden is traced through history from these Ancient Persian beginnings to the height of Mughal architecture (epitomised by the Taj Mahal), by way of the Muslim expansion through Central Asia and Europe. While the core elements of garden design were set in Ancient Persian times, and recur throughout the period studied, the impact of Islam on the local Persian culture brought about a new development of allegorical meaning associated with the garden. This allegorical development reached its apex, too, in the Taj Mahal in which, it is argued, the metaphorical representation of paradise in the garden tomb was made astonishingly explicit. The research for this mini thesis was gathered from secondary sources, including many published books and academic papers, photographic and diagrammatic evidence of extant ancient gardens, and reproductions of carpet designs.

These principles are, however, constantly challenged by the developments of e-commerce. This thesis considers the taxing of a permanent establishment and the influence of e-commerce on the concept of a permanent establishment. In 2000, the Organisation for Economic Co-operation and Development (“OECD”) developed and introduced guidelines on how to deal with e-commerce in the context of a permanent establishment. Since the OECD guidelines on e-commerce were issued, the permanent establishment principles have come under further scrutiny. The latest development came about in 2013 with the release of the Base Erosion and Profit Shifting (“BEPS”) Action Plan. This Action Plan addresses the intention of the OECD to deal with the taxing of the digital economy. With the development of e-commerce and the result of e-commerce creating intangible boundaries between countries, the concept of a virtual permanent establishment has emerged. This has resulted in the need to tax a presence of an enterprise in a jurisdiction where no actual physical connection can be established. Various authors have made suggestions on how to ensure that an economy in which business is being carried on is correctly compensated for in the form of taxes. The source of income is the driving force for the imposition of taxation today. The main goal of this thesis was to explore the alignment of the concepts of a permanent establishment and e-commerce in the digital economy. - Description: Current theories in geographical education advocate the use of strategies that encourage the pupil to play an active role in learning, thereby making such learning more meaningful and effective. Fieldwork is perceived as one such method. Fieldwork helps pupils acquire and develop understanding of geographical concepts, skills, attitudes and values through their own efforts and involvement. Fieldwork approaches have tended to change with the changing paradigms resulting in the development of approaches that are more pupil and experience oriented. In the South African school geography curricula fieldwork has been explicit since 1985. However, research has shown that in most South African secondary schools fieldwork as a teaching strategy is only applied to a limited extent. Teachers have always used financial constraints and time limitations as explanations for their failure to use fieldwork in teaching geography. This study attempts to demonstrate how the local environment of any school can be used effectively for teaching and learning most aspects of the senior secondary school geography syllabus, thereby alleviating the problems of time and money perceived by teachers as the major constraints inhibiting their use of fieldwork. In order to illustrate the effectiveness of fieldwork in the local environment, this study incorporated an analysis of the current senior secondary school geography syllabus, the identification of potential fieldwork sites in the Umtata District and the development and implementation of three fieldwork units based on three of the sites identified. The analysis of the evaluations of the three fieldwork units by the researcher, the pupils and the non-participant observer revealed that fieldwork conducted in the local environment is highly effective, interesting and rewarding to pupils even when they have no prior experience of fieldwork.

- Description: Current theories in geographical education advocate the use of strategies that encourage the pupil to play an active role in learning, thereby making such learning more meaningful and effective. Fieldwork is perceived as one such method. Fieldwork helps pupils acquire and develop understanding of geographical concepts, skills, attitudes and values through their own efforts and involvement. Fieldwork approaches have tended to change with the changing paradigms resulting in the development of approaches that are more pupil and experience oriented. In the South African school geography curricula fieldwork has been explicit since 1985. However, research has shown that in most South African secondary schools fieldwork as a teaching strategy is only applied to a limited extent. Teachers have always used financial constraints and time limitations as explanations for their failure to use fieldwork in teaching geography. The relationship should include non-bitterness, non-vengeance, unconditional love and respect for their human rights. Another significant dimension of the structure of forgiveness is the fact that the desire for the truth is not abandoned although forgiveness has taken place. Forgiveness also does not take away the effects of the violation. This means that one does not forget although forgiveness has been granted. Results further indicate that the forgiveness process is highly complex, individualized and not instantaneous. The individuals have to deal with various intrapersonal conflicts and anxieties as a result of the violation, before forgiveness is explored as an option, and before they can finally forgive. Significant interrelations between the structure and process of forgiveness were also identified.

Abstract Curriculum 2005 premised on Outcomes-Based Education is the new curriculum framework for South Africa. It signifies a paradigm shift in education from the traditional 'telling-listening' relationship between the teacher and the learner to one that emphasises leamer-centred approach to the teaching process. Teachers, though recognized as crucial to the educational transformation process in the country have also being identified as ill-equipped to meet the challenges posed by Outcomes-Based Education. This study starts with a brief overview of the South African curriculum and the main tenets underlying Outcomes-Based Education. The institutional conditions and whether the lecturers at a college in the Eastern Cape province perceive the need for a change in their classroom practice were also investigated. Bearing in mind the need for further research to validate the findings of this study, positive indicators that emerged from the study suggest the readiness of the college to implement Outcomes-Based Education at the proposed Education Development Centre.

- Description: This thesis presents a detailed chemical and spectroscopic determination of the capsular, polysaccharide K-antigen isolated from the Klebsiella bacterium, serotype K43 (culture #2482). The repeating unit of the capsular polysaccharide was found to be of the "3 + 2" repeating unit type. A uronic acid was found as part of a disaccharide side chain and the main chain of the polysaccharide was found to be composed of a neutral trisaccharide of mannose and galactose. The work forms part of an ongoing research interest in bacterial polysaccharides of this laboratory and now completes the structural elucidation of all the Klebsiella K-antigens, bar three antigens which were originally assigned to other laboratories. These data together with the respective serological characteristics of each serotype are available to the molecular biologist, and may result in the production of: vaccine(s) against Klebsiella infections, diagnostic products and novel carrier molecules enabling targeted drug delivery.

- Description: Changes in chlorophyll content, carotenoid content and composition, abscisic acid and phaseic acid levels, hydrolytic enzyme activity and polypeptide pattern were monitored during senescence of the primary attached leaves of Hordeum vulgare L. cv. Dyan. Senescence occurred due to the normal course of leaf development or was induced by incubation of leaves in darkness. Loss of chlorophyll and total leaf protein was retarded in light whereas it continued rapidly in leaves from dark-incubated seedlings. Chlorophyll alb ratio increased with the progression of senescence, suggesting that chlorophyll b was referentially degraded during this process. Loss of total protein coincided with enhanced activity of acid and neutral proteases. In contrast, loss of chlorophyll was not accompanied by an increase in· peroxidase activity, suggesting that this enzyme was not responsible for initiating chlorophyll breakdown. Carotenoid and abscisic acid levels were monitored in the same tissue extracts. The results obtained show that the increase in endogenous levels of abscisic acid, induced by senescence, correlated with enhanced epoxidation of the xanthophyll cycle, ie., increased conversion of zeaxanthin to antheraxanthin and all-trans-violaxanthin. In addition, an increase in abscisic acid levels occurred concomitant with a decrease in all-trans-violaxanthin and 9'-cis-neoxanthin, suggesting an apparent 1:1 relationship on a molar basis. It is therefore proposed that enhanced abscisic acid production, due to foliar senescence, arises from fluctuations in carotenoid turnover. Polypeptide patterns in isolated chloroplasts, purified thylakoid and stromal fractions were very similar for leaves incubated in either light or darkness. A decrease in intensity of bands was observed in isolated chloroplasts and stromal fractions. Intensity of bands in thylakoids remained unchanged with the progression of senescence. Protein standards of peroxidase and lipoxygenase co-migrated with proteins of the isolated chloroplast. Although tentative, some proteins of the chloroplast may be representative of precursors of hydrolytic enzymes which are known to increase during senescence.

- Description: Consider, for instance, you and your family have lived around a place where you enjoyed the flora and fauna of the land as well as the natural environment. Fishing and farming were pleasant activities for your family, and anyone in the community not only to survive but thrive. Your neighbours and the people in the community pride themselves so much in their farming abilities and fishing techniques. Suddenly things start taking a different turn because of rising sea level and changing weather pattern. First, your land began to give way because of sweeping erosion, and later the riverbank serving the community starts drying up. Your neighbour could not hold their own in this grim condition as they started relocating. You even witnessed some of your family and friends leaving the community and you later heard that they have become climate refugees. To put it all together, you were told that what you are experiencing is one of climate change effects. You were told that the loss of the place that you once cherished is not an act of God but the result of the reckless flaring of greenhouse gasses harmful to the environment by some powerful but polluting nations. What will you do? This might sound like a fairy tale. In reality, this is the story of the people of Saint Louis Atlantic Coasts in Senegal, captured by the Global Climate Change Alliance Plus Initiative.1 Their situation has been described as ‘living on the edge’ because for centuries, Saint Louis Atlantic Coasts, home to around 230,000 inhabitants, also known as the ‘Venice of Africa’ has been protected from the pounding Atlantic waves by the ‘Langue de Barbarie,’ a narrow, 30 km peninsula at the mouth of the Senegal river. However, the low-lying sandy spit of land along this World Heritage site is itself rapidly disappearing in the present. This is the consequence of a changing climate and other man-made problems such as illegal sand-mining and over-development. Today, the southern part of the Langue de Barbarie is an island, and the village of Doune Baba Dieye is under more than a metre of water. The villagers have become climate refugees, forced to live in temporary camps on the mainland. Not only have they lost their homes, but they have also lost significant cultural heritage like the farming and fishing culture tied to this place. The ethical considerations that emerge from climate change impacts on the world’s cultural heritage are varied. However, it seems not as self-evident in the way that research on climate change ethics has been framed around economic interest and direct threats to human life and other species. Even when they mention climate impacts on heritage sites around the world, those of Africans have been side-lined. For instance, the impact of climate change on small island nations like Tuvalu, Kiribati2 have gained much traction in climate change discourse, but we do not get to see stories of Africans whose cherished cultural heritage are affected by climate despoliation. How do we respond to this intractable challenge? This is a question of justice and, to be more precise, climate justice. Many principles and proposals for climate justice have been put forward, but the insufficient attention to the vulnerabilities or loss of cultural heritage values of Africans, which is a critical aspect of their social realities, make these theories less persuasive on a global level. This thesis, then, fills this gap in the literature by suggesting that the failure to take cognizance of the injustice in neglecting cultural heritage values when dealing with the burden of climate change is the effect of three problems. One, the value of culture is less understood in this environmental age. Hence, cultural values are excluded or made to be secondary in consideration of principles of climate justice. Second, the idea of Personhood has been neglected in climate ethics and climate justice discourse. Yet, this idea of Personhood can be an enabler of climate justice in that a realization of the significance of cultural heritage to the wellness of the human persons in Africa, makes it loss morally reprehensible. Third, those whose cultural heritage is significantly affected do not get represented in the debate about sharing the burden of climate change. This dissertation thereby builds upon the general findings of the past about anthropogenic climate change, its causes and consequences. Adopting a discursive normative framework, I also address the significance of cultural heritage in this contemporary environmental age and discuss the global justice implications of cultural heritage loss to climate change. This dissertation further provides a critique of mainstream climate justice theories, especially their marginalization of the cultural dimension of climate change. In this regard, the metaphor - ‘cultural storm’ was deployed to argue that climate justice discourses have neither factored the deep socio-cultural impact of climate change nor do they draw on the cultural understanding of justice in putting forward their theories. Given the nature of the indirect, cumulative, and interconnected invisible losses to cultural heritage from climate change, it seems unlikely that they can be addressed by simple tweaks of the climate justice status quo. This dissertation proposes that the idea of personhood in African philosophy, can be conceived to ensure climate justice live up to its expectations in a world of diverse persons dealing with a complex phenomenon like climate change. It argues that a cultural dimension of climate ethics has implications for how mitigation, adaptation and compensation plans should be furthered for global climate justice. At the foundation of my argument, I suggest that what is needed in climate justice discourse is a commitment to explore new and innovative alternatives that will produce an inclusive global climate treaty that is sensitive to the cultural heritage assets that is destroyed by climate change in Africa. This will require a multi-dimensional framework that allows fundamentally different kinds of values and benefits to be given equal visibility and standing in global climate negotiations. The dissertation proceeds in six chapters. In the first chapter, I discuss how climate change denies, damage and destroys cultural heritage values in Africa and argue that it is unjust to ignore this dimension of climate change impact, particularly on the African continent. In the second chapter, I critically discuss the normative value of cultural heritage in an environmentally sustainable and morally appropriate way for this global age. I argue that what is lost when climate change affects cultural heritage is a significant cultural asset that ought to be seriously considered in climate ethics. The third chapter addresses the global justice implications of the destruction of Africa's cultural heritage by climate change that must be paid attention to. In chapter four, I evaluate the plausibility of some of the mainstream climate justice proposals. I offered a different possible critique of current approaches to climate justice to show how they have furthered cultural injustice. The critique of current climate justice theories that I offer, stems from an uninspiring approach that belies logic permitting the sacrificing of that which is connected to others wellbeing as well as the implicit assumptions and the limitation of the idea of justice that undergird these climate justice theories. - Description: Consider, for instance, you and your family have lived around a place where you enjoyed the flora and fauna of the land as well as the natural environment. Fishing and farming were pleasant activities for your family, and anyone in the community not only to survive but thrive. Your neighbours and the people in the community pride themselves so much in their farming abilities and fishing techniques. Suddenly things start taking a different turn because of rising sea level and changing weather pattern. First, your land began to give way because of sweeping erosion, and later the riverbank serving the community starts drying up. Your neighbour could not hold their own in this grim condition as they started relocating. You even witnessed some of your family and friends leaving the community and you later heard that they have become climate refugees. To put it all together, you were told that what you are experiencing is one of climate change effects. You were told that the loss of the place that you once cherished is not an act of God but the result of the reckless flaring of greenhouse gasses harmful to the environment by some powerful but polluting nations. What will you do? This might sound like a fairy tale. In reality, this is the story of the people of Saint Louis Atlantic Coasts in Senegal, captured by the Global Climate Change Alliance Plus Initiative.1 Their situation has been described as ‘living on the edge’ because for centuries, Saint Louis Atlantic Coasts, home to around 230,000 inhabitants, also known as the ‘Venice of Africa’ has been protected from the pounding Atlantic waves by the ‘Langue de Barbarie,’ a narrow, 30 km peninsula at the mouth of the Senegal river. However, the low-lying sandy spit of land along this World Heritage site is itself rapidly disappearing in the present. This is the consequence of a changing climate and other man-made problems such as illegal sand-mining and over-development. Today, the southern part of the Langue de Barbarie is an island, and the village of Doune Baba Dieye is under more than a metre of water. The villagers have become climate refugees, forced to live in temporary camps on the mainland. Not only have they lost their homes, but they have also lost significant cultural heritage like the farming and fishing culture tied to this place. The ethical considerations that emerge from climate change impacts on the world’s cultural heritage are varied. However, it seems not as self-evident in the way that research on climate change ethics has been framed around economic interest and direct threats to human life and other species. Even when they mention climate impacts on heritage sites around the world, those of Africans have been side-lined. For instance, the impact of climate change on small island nations like Tuvalu, Kiribati2 have gained much traction in climate change discourse, but we do not get to see stories of Africans whose cherished cultural heritage are affected by climate despoliation. How do we respond to this intractable challenge? This is a question of justice and, to be more precise, climate justice. Many principles and proposals for climate justice have been put forward, but the insufficient attention to the vulnerabilities or loss of cultural heritage values of Africans, which is a critical aspect of their social realities, make these theories less persuasive on a global level. This thesis, then, fills this gap in the literature by suggesting that the failure to take cognizance of the injustice in neglecting cultural heritage values when dealing with the burden of climate change is the effect of three problems. One, the value of culture is less understood in this environmental age. Hence, cultural values are excluded or made to be secondary in consideration of principles of climate justice. Second, the idea of Personhood has been neglected in climate ethics and climate justice discourse. Yet, this idea of Personhood can be an enabler of climate justice in that a realization of the significance of cultural heritage to the wellness of the human persons in Africa, makes it loss morally reprehensible. Third, those whose cultural heritage is significantly affected do not get represented in the debate about sharing the burden of climate change. This dissertation thereby builds upon the general findings of the past about anthropogenic climate change, its causes and consequences. Adopting a discursive normative framework, I also address the significance of cultural heritage in this contemporary environmental age and discuss the global justice implications of cultural heritage loss to climate change. This dissertation further provides a critique of mainstream climate justice theories, especially their marginalization of the cultural dimension of climate change. In this regard, the metaphor - ‘cultural storm’ was deployed to argue that climate justice discourses have neither factored the deep socio-cultural impact of climate change nor do they draw on the cultural understanding of justice in putting forward their theories. Given the nature of the indirect, cumulative, and interconnected invisible losses to cultural heritage from climate change, it seems unlikely that they can be addressed by simple tweaks of the climate justice status quo. This dissertation proposes that the idea of personhood in African philosophy, can be conceived to ensure climate justice live up to its expectations in a world of diverse persons dealing with a complex phenomenon like climate change. It argues that a cultural dimension of climate ethics has implications for how mitigation, adaptation and compensation plans should be furthered for global climate justice. At the foundation of my argument, I suggest that what is needed in climate justice discourse is a commitment to explore new and innovative alternatives that will produce an inclusive global climate treaty that is sensitive to the cultural heritage assets that is destroyed by climate change in Africa. This will require a multi-dimensional framework that allows fundamentally different kinds of values and benefits to be given equal visibility and standing in global climate negotiations. The dissertation proceeds in six chapters. In the first chapter, I discuss how climate change denies, damage and destroys cultural heritage values in Africa and argue that it is unjust to ignore this dimension of climate change impact, particularly on the African continent. In the second chapter, I critically discuss the normative value of cultural heritage in an environmentally sustainable and morally appropriate way for this global age. I argue that what is lost when climate change affects cultural heritage is a significant cultural asset that ought to be seriously considered in climate ethics. The third chapter addresses the global justice implications of the destruction of Africa's cultural heritage by climate change that must be paid attention to. In chapter four, I evaluate the plausibility of some of the mainstream climate justice proposals. I offered a different possible critique of current approaches to climate justice to show how they have furthered cultural injustice. The critique of current climate justice theories that I offer, stems from an uninspiring approach that belies logic permitting the sacrificing of that which is connected to others wellbeing as well as the implicit assumptions and the limitation of the idea of justice that undergird these climate justice theories. Although there are available treatments, their efficacy has been greatly reduced over the past two decades due to the development of resistance to currently available drugs. This has necessitated the search for new and effective antimalarial agents. This project approached the search for new antimalarial compounds in two ways: (i) by screening natural products isolated from marine algae against the Plasmodium parasite and (ii) by modification of selected isolated active compounds to target 1-deoxY-đ-xylulose 5-phosphate reductoisomerase (DXR), an enzyme found in the nonmevalonate isoprenoid biosynthetic pathway of Plasmodium Jalciparum. It was envisaged that such a compound would exhibit dual action on the Plasmodium parasite. Extracts obtained from 22 marine algae were prefractionated by solvent partitioning and were screened for anti plasmodial activity against the chloroquine sensitive (CQS) P. Jalciparum D 10 strain. Overall, 50% of the algae screened produced at least one crude fraction with activity against P. Jalciparum. Extracts of the algae Sargassum heterophyllum, Plocamium cornutum, Amphiroa ephedrea and Pterosiphonia cloiophylla gave the most promising results. Fractionation of S. heterophyllum afforded three tetraprenyltoluquinols (3.1, 3.2 and 3.5) and an all-trans-fucoxanthin (3.6). Three new compounds (4.5, 4.6 and 4.7) and two known halogenated monoterpenes (4.1 and 4.4) were isolated from P. cornutum. Each of the isolated compounds from both S. heterophyllum and P. cornutum showed antiplasmodial activity with IC₅₀ values ranging from 2.0 - 15.3 μM for S. heterophyllum and 13 - 230 μM for P. cornutum. Attempts to synthetically modify halogenated monoterpene 4.4 by dihydroxylation and phosphorylation in order to inhibit the DXR enzyme was unsuccessful. However, the hemiterpene analogue (5.42) of the halogenated monoterpenes was successfully phosphorylated and dihydroxylated to give compound 5.45 which showed promising activity against DXR. The result obtained indicated that the proposed phosphorylation and dihydroxylation of the halogenated monoterpene 4.4 would result in the synthesis of a potent DXR inhibitor and therefore a potential antimalarial agent with dual mode of action on the Plasmodium parasite.

- Description: Malaria continues to be a devastating disease and a major cause of death in sub-Saharan Africa. With resistance against most of the available antimalarial drugs, there is a need for ongoing research and development of antimalarial agents. Fosmidomycin and its acetyl analogue FR900098 have been identified as potent inhibitors of Plasmodium falciparum, the causative agent of the most deadly form of malaria. Clinical trials of these agents have revealed poor absorption due to their high hydrophilicity. In the present studies the effect of halogenation of the acyl chain as well as the biological effect of extending the acyl sidechain was explored. This provided the basis on which fosmidomycin hybrids were designed to investigate the feasibility of hybrid extending into NADPH binding pocket. Synthesis of a series of halogenated FR900098 analogues was carried out in three stages. This included i) The introduction of the phosphonate group by reaction with 1,3dibromopropane in an Arbuzov reaction, ii) The introduction of a hydroxamate group by reaction of the propyl phosphonate by means of a nucleophilic substitution reaction with BocNHOBn and iii) The introduction of a halogenated acyl side chain on a protected fosmidomycin backbone. The synthesis of fosmidomycin-hybrids for which chloroquinefosmidomycin hybrids were used as the prototype, involved convergence of the two separately constructed moieties i.e. fosmidomycin and the quinoline moieties in a covalent linkage. The quinoline moiety was easily synthesized from the reaction of 4,7dichloroquinoline with 1,2-diamino ethane. The aminoquinoline so formed resulted in chloroquine-fosmidomycin hybrids 3.8 and 3.9 when reacted with halogenated FR900098 analogues. Antiplasmodial assays were conducted on the chloroquine-fosmidomycin hybrids and the halogenated fosmidomycin derivatives against the chloroquine resistant Gambian FCR-3 strain of P. falciparum. The most potent iodoacetyl fosmidomycin analogues 2.21 gave an IC50 value of 5.54 µM which is eight times more potent than the known antiplasmodial FR900098 which gave an IC50 value of 41.67 µM. All the halogenated FR900098 analogues showed better antiplasmodial activity than their non-halogenated derivatives. This indicated that the presence of halogens in the FR900098 analogues contributes to their biological Chapter 1 Literature review activity. The acetyl and propyl linked hybrids 3.8 and 3.9 showed potent antiplasmodial activity with IC50 values of 0.18 and 0.82 µM respectively. These were by far the most potent hybrids synthesized and provided leads for a new class of promising antimalarial agents. Preliminary E. coli DXR enzyme inhibition assays were carried out on the halogenated fosmidomycin analogues. The results showed good inhibition of the enzyme by the phosphonic acids of the chloroacetyl and chloropropyl analogues 2.1 and 2.2 respectively. Molecular modelling of the compounds on E. coli (PDB code: 2EGH) and P. falciparum (PDB code: 3AUA) DXR showed strong binding of the halogenated fosmidomycin analogues while the hybrids in the absence of docked NADPH showed minimum binding to the enzymes.

- Description: Plasmodium falciparum is a protozoan parasite that causes a severe form of malaria, a mosquito-borne infectious disease in humans. P. falciparum encodes a number of proteins to facilitate its life-cycle, including a type II heat shock protein 40 (Hsp40), Pfj2. Pfj2 shows a degree of homology to human ERdj5, a resident protein of the endoplasmic reticulum (ER) that promotes protein quality control by facilitating the degradation of misfolded proteins. The overall aim of this study was to further understand the function of Pfj2 in the P. falciparum cell by characterising it biochemically. A bioinformatic analysis of Pfj2 was carried out to enable the identification of a potential ER signal sequence and cleavage site. Furthermore, an analysis of Pfj2 protein sequence was performed to compare domain similarities and identities with typical type II Hsp40s namely, human ERdj5, S. cerevisiae Sis1, human Hsj1a and human DnaJB4. The method used included the insertion of the codon-optimised coding sequence for the processed ER form of Pfj2 into the prokaryotic expression vector, pQE30, to enable overproduction of a histidine-tagged protein. A 62 kDa His₆-Pfj2 was successfully expressed in Escherichia coli and purified using denaturing nickel affinity chromatography. ATPase assays were performed to determine the ability of His₆- Pfj2 to stimulate the chaperone activity of the ER Hsp70, also called immunoglobulin binding protein (BiP). Initial studies were conducted on readily available mammalian His₆-BiP as a control, which was shown to have an intrinsic activity of 12.07±3.92 nmolPi/min/mg. His₆- Pfj2 did not stimulate the ATPase activity of mammalian His₆-BiP, suggesting that it either could not act as a co-chaperone of mammalian His₆-BiP (specificity), or it required a misfolded substrate in the system. Therefore, ongoing studies are addressing the interaction of Pfj2 and misfolded substrates with P. falciparum BiP. The results of these studies will further our understanding of a poorly-studied parasite chaperone that represents a potential drug target for development of novel strategies for the control of a serious human disease

This commercial oriented journalism gives priority to articles that attract mass audiences like entertainment while it downplays information that promotes debates that is necessary for citizens to be able to have a voice on the issues that affect them. And since participation and discussion are cornerstones of a democratic process, market-driven journalism undermines democracy because it narrows down the forum for debate. As a result, active citizens are turned into passive observers in society. Although several studies about this phenomenon have been done in the western world, the same is happening in Africa because the media face similar challenges as in the West; challenges of globalisation and media conglomeration facilitated by the rapid advancing technology. This study, which is informed by political economy and market-driven journalism theories, notes that the media in Uganda are also faced with these challenges. The study is focused on Uganda’s only independent newspaper, The Monitor. The findings indicate that market-driven journalism is taking root at the expense of journalism that promotes citizenship and debate such as political reporting and opinions. For example, there has been an increase of entertainment, sports and supplement articles in The Monitor as compared to declining political reporting and opinions. More so, investigative reporting has dwindled over the years at the expense of increasing use of press releases. This research was conducted within the context of the food services sector of Higher Education Institution dining halls and in the midst of the rising global call for food resource management and food waste reduction. The main aim of this research therefore was to identify opportunities for learning and change for a more sustainable food economy, contributing to Education for Sustainable Production and Consumption, and by further implication, Education for Sustainable Development. To achieve this aim, I used Cultural Historical Activity Theory as theoretical and methodological framework; drawing on the second and the third generations of this theory. Implicated in the above research approach is the identification of expansive learning opportunities from the surfacing of ‘tensions’ and ‘contradictions’. In this case study of the Rhodes University Campus Food Services, such tensions and contradictions inhibiting a more sustainable food economy, involving food waste production were identified. To narrow the scope of the study, one dining hall formed the focus of the case, with a two phased research approach whereby one research question and three goals were developed for each phase. The former being the exploration phase and the latter being the initial stages of the expansive phase. Methods used in line with the methodological framework included ten individual interviews with food producers (staff members), nine focus group discussions with food consumers (students), observations of the dining hall activities which lasted for over a month and two ‘Change Laboratory Workshops’. Some of the findings of this research are that food wastage cannot be addressed and appropriately curtailed without an intensive consideration of all the stages of food economy. Multiple contradictions and sources of tensions embedded in the Food Services Sector constituted major causes of food waste. Additionally, the lack of substantial food waste related teaching and learning activities, the presence of disputed rules, institutional structure and traditional practices within the Food Services all exacerbated the tensions and contradictions. More so, prioritizing some of this identified contradictions and tensions hindering a more sustainable food economy and relegating some as unimportant or nonurgent is unproductive. Finally, the non-existence of facilitated deliberation, consultation, dialogue, collaboration between food producers and food consumers has been identified as an obstacle to learning and institutional change. Recommendations abound in re-orienting, re-educating, and re-informing the constituents of the food economy. Re-visiting and revising of rules and regulations guiding conduct of students and kitchen staff members in the RU dining halls, as well as revision of existing learning support materials and mediating tools in use is needed. Recognition and consideration of the concerns and interests of students and kitchen staff members are also needed. Finally, there is a need to continue to address the tensions and contradictions identified in this case study, to further the Expansive Learning Process if a more sustainable food economy at Rhodes University is to be established.

A morphogenic and laminated system explanation of position-practice systems and professional development training in mainstreaming education for sustainable development in African universities

It explores the roles and practices of these institutions, especially their professionals, in the Anthropocene era where increasing concern for contemporary environmental and sustainability issues and risks emerge. The study presents a longitudinal case study of institutions and participants of the Swedish/African/Asian International Training Programme (ITP) on ESD in Higher Education (HE), who are mostly university educators. This thesis however focuses on African ITP participants only. At a macro level, the research sought to examine how African university educators have contributed to the United Nations Decade of Education for Sustainable Development (UNDESD) through their participation in the ITP (which is a change oriented professional development training programme on ESD) and the associated ESD ‘change projects’. The change projects are ITP participants’ direct attempts to mainstream environment and sustainability issues, concerns, and concepts into core university functions and practices: teaching, research, community engagement, and management operations and policy engagement. At a meso level the study sought insight into how educators in national institutions were supported by sub-regional and regional initiatives, institutions and organisations, including the Mainstreaming of Environment and Sustainability in African (MESA) Universities Partnership programme, especially an initiative supported by the Southern African Development Community Regional Environmental Education Programme to provide (limited) seed funding to three southern African universities to establish what are known as ‘MESA Chairs’, with dedicated time and support for MESA activities in their universities . At a micro level, this research sought to investigate how the position-practice systems and the ITP shape (enable or constrain) effective ESD mainstreaming in higher education, and how the morphogenetic approach and laminated system can be used to understand and explain these dynamics and their relations with meso and macro level engagements. The research sought to understand these dynamics through empirical investigations using survey questionnaires, interviews, document analysis and field visits. The research is constituted as theoretical, conceptual, methodological and analytical exploration using a singular and nested case study research approach, underlaboured by a critical realist ontology, and drawing on a social learning epistemology and social realist morphogenetic interpretive lens. In particular, ontological depth was sought via critical realist laminated system explanation. See Chapter Two for details. This study was carried out in three phases. Phase one encapsulates the investigation of all ITP ESD in higher education alumni who were Asian and African participants from the inception of the ITP to its completion, over a six-year period (2008-2013). This included 280 academics from Asia and Africa in 35 countries in Asia and Africa from 106 institutions in Asia and Africa with their 139 change projects. The outcome of phase one of the research is only included in this thesis as an appendix (see Appendix 3; Agbedahin & Lotz-Sisitka, 2015). However, this phase provided and formed the foundational data that was expanded in phases two and three for the purpose of this study. Phase two of this research concentrated on a less broad population of research participants comprising only all African ITP alumni, from all regions in Africa. The overall data collection and analysis included 162 academics in 23 African countries from 66 institutions with their 81 change projects. The aim was to investigate and provide a morphogenetic explanation of their change projects and how the relationship between participants’ positions and practices (and that of others) may influence ESD mainstreaming in universities. The outcome of this phase two investigation is presented in Chapter Four. In phase three, (nested) case studies of Swaziland, Zambia, and Botswana (in the southern Africa region), which included all the ESD ITP HE participants therein and the three corresponding EE/ESD MESA Chairs, were developed. The population sample in this phase three therefore contained 20 academics, from six institutions with their nine change projects. This phase was characterised by field trips to these countries and in-depth data collection and analysis in order to investigate and deepen the morphogenetic explanations of their change projects and how the relationship between participants’ positions and practices (and that of others) have indeed influenced the ESD mainstreaming in universities. The outcome of the theoretical exploration underpinning this study provided a potential model for understanding ESD learning and change processes that are facilitated by professional development training programmes in the context of ESD in HE. This study also provides a model for appraising educational changes in time and in space, especially in relation to ESD, or the types of changes that can be brought about by professional development interventions such as those provided by the ITP and how they can be tracked, monitored and documented. For the field of professional or academic development in higher education, this research highlights the significance of the relationship between position-practice systems, professional development interventions and institutional transformation. For the field of ESD in higher education, this study shows the need for in-depth consideration of the position-practice system and sphere of influence of change agents and related stakeholders in and around their institutions in the design and development of professional development programmes. - Description: This thesis examines the phenomenon of child-soldiering from a different perspective. It seeks to challenge, using a novel approach, earlier studies on the roles of former child-soldiers in post-war societies. It focuses on the subjectivity of young veterans, that is war veterans formerly associated with armed forces and groups as children during the 14-year gruesome civil war which bedevilled Liberia between 1989 and 2003. This civil war claimed roughly 250,000 lives, and saw the active participation of approximately 21,000 child-soldiers. This thesis departs from previous works which mostly painted an apocalyptic picture of young veterans, and explores the nexus between their self-agency, Foucauldian technologies of the self and their transformation in the post-war society. The majority of previous scholarly works which have dominated the field of child-soldiering dwelt on the impact of armed conflict on the child-soldiers, the negative consequences, the causes of child-soldiering, and the rehabilitation and reintegration of the young veterans after their disarmament and demobilization. What this thesis seeks to do however, is to establish that, rather than considering the young veterans simply as social misfits, distraught and dispirited human beings, it should be noted that young veterans through their agency, are capable of ensuring their reintegration into their war-ravaged societies. Sadly, these young former fighters’ self-agency and technologies of the self in defining their civilian trajectories have often been overshadowed by vaunted humanitarian aid and multilayered war-profiteering. This study is underpinned by interpretive constructivism, symbolic interactionism, social identity theory, sociometer theory and expectancy theory, and sheds light on how young veterans’ self-agency, instrumental coalitions, and decision-making processes, synergistically shifted the negative identities foisted on them as a result of their participation in the war.

- Description: This thesis examines the phenomenon of child-soldiering from a different perspective. It seeks to challenge, using a novel approach, earlier studies on the roles of former child-soldiers in post-war societies. It focuses on the subjectivity of young veterans, that is war veterans formerly associated with armed forces and groups as children during the 14-year gruesome civil war which bedevilled Liberia between 1989 and 2003. This civil war claimed roughly 250,000 lives, and saw the active participation of approximately 21,000 child-soldiers. This thesis departs from previous works which mostly painted an apocalyptic picture of young veterans, and explores the nexus between their self-agency, Foucauldian technologies of the self and their transformation in the post-war society. - Description: Challenges of access to education in the developing world and elsewhere appear to be widespread. Many declarations and conventions have been developed to assist countries to respond to the issue. While challenges of access are universal, Africa in general and sub-Saharan Africa in particular are presented with additional concerns about gender equality and gender parity. While learner numbers seem to be on the decline globally, dropout amongst girls is disproportionately greater than amongst boys. Even though school retention presents a challenge at all levels of the schooling system, it is more acute for girls at the secondary school level. This study was conducted to examine and understand factors that shape retention of secondary school female learners in two rural public schools in Alimosho Region of Lagos State, Nigeria. The study design was qualitative and interpretive in nature. Data collection strategies included administered questionnaires in two schools, focus group discussion with twenty female learners in two schools, case studies, individual interviews with four participants from two schools, and observation in English and Biology classes where the two teachers from the two schools participated in the interviews. Ethical clearance from Alimosho Educational Region office and the two schools was obtained before undertaking the study. Participants’ school principals also signed written consent forms before interviews. The female learners were briefed about the study interview activities and advised that their participation was voluntary and that they were free to withdraw at any point. This study drew on Sen’s (1989, 2000) capabilities theory to understand the phenomenon beyond dominant discourses on education that view education as a basic human right or that focus on economic and development gains. The key finding of this study is that in-school and out- of- school factors interact in complex ways to support female learner retention. Key among these are value placed on education by female learners and significant others, particularly parents; family support; and individual aspirations. Extra-curricular participation, government policy, role models, and peer support were also found to be important factors that mediate progression and retention.
