Spatial structure of recruitment in the mussel Perna perna at local scales: effects of adults, algae and recruit size

Spatial heterogeneity was estimated by analyzing scaling properties of semivariograms using a fractal approach. Relationships between density of mussel recruits and adults and biomass of the red alga Gelidium pristoides at the different scales were examined by cross-semivariograms. We found that the distribution of adults showed spatial dependence at all transects, often with higher spatial heterogeneity (higher fractal dimension, D) at smaller scales (1st scaling region). The distribution of larger recruits exhibited spatial dependence at all transects, revealing a spatial structure, which was related to that of adults. In contrast, the distribution of late plantigrades showed mainly spatial independence (random pattern; 1.97 < D ≤ 2). Densities of both size classes of recruits were positively related to those of adults at all transects and scales, but the relationship was stronger for larger recruits than late plantigrades, explaining why there was clearer spatial structure of larger recruits. The relationship with algae was mainly negative for larger recruits, while it tended to be positive at many scales for late plantigrades. Thus, both adult mussels and G. pristoides are suitable habitats for plantigrades, while mussels are the main habitat for larger recruits. This may mean that recruits on algae either die or migrate to mussel clumps at a certain size. This study highlights the importance of recruit size when analyzing recruitment patchiness of mussels, and has implications for sustainable management of P. perna.

The combined grazing impact of the two copepods ranged between 0.65 and 4.37 mg chl-a m^(–3), or between 4.3% and 35.9% of the available chl-a in the water column. Variations in the grazing activity of the two species could be attributed largely to seasonality in water temperature and shifts in the phytoplankton community structure and zooplankton abundance.

- Description: A detailed hydrographic and biological survey was carried out in the region of the South-West Indian Ridge during April 2004. Altimetry and hydrographic data have identified this region as an area of high flow variability. Hydrographic data revealed that here the Subantarctic Polar Front (SAF) and Antarctic Polar Front (APF) converge to form a highly intense frontal system. Water masses identified during the survey showed a distinct separation in properties between the northwestern and southeastern corners. In the north-west, water masses were distinctly Subantarctic (>8.5°C, salinity >34.2), suggesting that the SAF lay extremely far to the south. In the southeast corner water masses were typical of the Antarctic zone, showing a distinct subsurface temperature minimum of <2.5°C. Total integrated chl-a concentration during the survey ranged from 4.15 to 22.81 mg chl-a m[superscript (-2)], with the highest concentrations recorded at stations occupied in the frontal region. These data suggest that the region of the South-West Indian Ridge represents not only an area of elevated biological activity but also acts as a strong biogeographic barrier to the spatial distribution of zooplankton.

Lack of variation within and between east and west coast samples precluded further population genetic analysis. The sequence obtained in this study was also compared with other sequences lodged in the Genbank database. Phylogenetically, the South African O. vulgaris is closely related to O. vulgaris from Senegal (0.67% divergence) and the Mediterranean (1.51% divergence). Within the Mediterranean group, O. vulgaris from South Africa displayed less sequence divergence from Senegalese and Mediterranean individuals than O. vulgaris from Venezuela (3.85%) and Taiwan (3.87%). These data do not, therefore, refute the hypothesis of a single O. vulgaris genetic population around the coast.

- Description: Little is known about pollination of the Aizoaceae (Mesembryanthemaceae). There are sparse reports of generalist pollination in the family by a variety of insects (predominantly bees). Furthermore, most species are self-incompatible in cultivation. In this study, observations were made on two populations of Bergeranthus multiceps (Salm-Dyck) Schwantes growing in the Eastern Cape province of South Africa. Insects visiting the flowers were collected and examined for pollen. While 79 individual insects (in 24 genera representing 14 families and four orders) were collected visiting the flowers, the majority (43 individuals) were female Allodapula variegata bees (Apidae, subfamily Xylocopinae, tribe Allodapini)collecting pollen. All other bee visitors were also female, suggesting pollen collection as the primary activity at the flowers. The protandrous flowers were found to be self-incompatible, pointing to the importance of bee-mediated xenogamy in this species. The flowers of B. multiceps are bright yellow in the human visual spectrum. In addition, the petals of this species reflect ultraviolet light. In contrast, the yellow anthers absorb UV. Flower opening and closing is common in the Aizoaceae. Interestingly, in B. multiceps flowers open at about 15:30 and remain open for approximately three hours before closing again in the late afternoon. These afternoon flower opening events were found to be closely correlated to ambient temperatures above 23°C, relative humidity lower than 50% and vapour pressure deficit below 1.05 kPa measured from as early as 09:00 on the days when flowers opened.

Additional taxonomic problems concerning the possible segregation of Podocarpus milanjianus from P. latifolius are addressed using DNA sequence data from the nuclear internal transcribed spacer 2 (ITS2) region. - Description: Leucadendron is a moderately large genus of Proteaceae almost entirely restricted to the Cape Floristic Region of southern Africa. The genus is unusual in being dioecious and sexually dimorphic. ITS sequence data were obtained from 62 of the 96 currently recognized taxa (85 species and 11 subspecies). Phylogenetic analyses were conducted under Maximum Likelihood and parsimony and resolved nine groups of species with varying degrees of bootstrap support, but relationships between these groups are largely unsupported. The phylogeny conflicts with the current taxonomic arrangement, which is based mainly on fruit morphology. The two sections of the genus, Alatosperma and Leucadendron, and several subsections within these sections, are resolved as non-monophyletic. This means that taxonomically important characters (such as fruit shape) have evolved multiple times, as the species with nutlike fruit (resolved into two of the nine groups) appear to have evolved independently from ancestors with winged fruit. Based on the topology obtained, the life history traits of anemophily, myrmechochory, and re-sprouting have also originated multiple times. Dispersal–Vicariance (DIVA) analysis suggests that the genus had an ancestral area in the Karoo Mountain and Southeastern phytogeographic centres of endemism in the southwestern Cape. , Elsevier

- Description: Leucadendron is a moderately large genus of Proteaceae almost entirely restricted to the Cape Floristic Region of southern Africa. The genus is unusual in being dioecious and sexually dimorphic. ITS sequence data were obtained from 62 of the 96 currently recognized taxa (85 species and 11 subspecies). Phylogenetic analyses were conducted under Maximum Likelihood and parsimony and resolved nine groups of species with varying degrees of bootstrap support, but relationships between these groups are largely unsupported. The phylogeny conflicts with the current taxonomic arrangement, which is based mainly on fruit morphology. The two sections of the genus, Alatosperma and Leucadendron, and several subsections within these sections, are resolved as non-monophyletic. This means that taxonomically important characters (such as fruit shape) have evolved multiple times, as the species with nutlike fruit (resolved into two of the nine groups) appear to have evolved independently from ancestors with winged fruit. Based on the topology obtained, the life history traits of anemophily, myrmechochory, and re-sprouting have also originated multiple times. Dispersal–Vicariance (DIVA) analysis suggests that the genus had an ancestral area in the Karoo Mountain and Southeastern phytogeographic centres of endemism in the southwestern Cape. , Elsevier

We investigate endothermy as a thermoregulatory strategy in 11 South African species from five genera, with comparisons to the lone ectothermic platypleurine we found, in an attempt to ascertain any influence that habitat and/or body size have on the expression of endothermy in the platypleurine cicadas. Field measurements of body temperature (Tb) show that these animals regulate Tb through endogenous heat production. Heat production in the laboratory elevated Tb to the same range as in animals active in the field. Maximum Tb measured during calling activity when there was no access to solar radiation ranged from 13.2° to 22.3°C above ambient temperature in the five species measured. The mean Tb during activity without access to solar radiation did not differ from the mean Tb during diurnal activity. All platypleurines exhibit a unique behavior for cicadas while warming endogenously, a temperature- dependent telescoping pulsation of the abdomen that probably functions in ventilation. Platypleurines generally call from trunks and branches within the canopy and appear to rely on endothermy even when the sun is available to elevate Tb, in contrast to the facultative endothermy exhibited by New World endothermic species. The two exceptions to this generalization we found within the platypleurines are Platypleura wahlbergi and Albanycada albigera, which were the smallest species studied. The small size of P. wahlbergi appears to have altered their thermoregulatory strategy to one of facultative endothermy, whereby they use the sun when it is available to facilitate increases in Tb. Albanycada albigera is the only ectothermic platypleurine we found. The habitat and host plant association of A. albigera appear to have influenced the choice of ectothermy as a thermoregulatory strategy, as the species possesses the metabolic machinery to elevate to the Tb range observed in the endothermic species. Therefore, size and habitat appear to influence the expression of thermoregulatory strategies in African platypleurine cicadas.

The technique can be used to predict potential distributions of organisms using presence-only locality records and a set of environmental predictor variables. FEM uses fuzzy logic to classify a set of predictor variable maps based on the values associated with presence records and combines the results to produce a potential distribution map for a target species. This technique represents several refinements of the envelope approach used in the BIOCLIM modelling package. These refinements are related to the way in which FEMs deal with uncertainty, the way in which this uncertainty is represented in the resultant potential distribution maps, and the way that these maps can be interpreted and applied. To illustrate its potential use in biogeographical studies, FEM was applied to predicting the potential distribution of three invasive alien plant species (Lantana camara L., Ricinus communis L. and Solanum mauritianum Scop.), and three native cicada species (Capicada decora Germar, Platypleura deusta Thun. and P. capensis L.) in South Africa, Lesotho and Swaziland. These models were quantitatively compared with models produced by means of the algorithm used in the BIOCLIM modelling package, which is referred to as a crisp envelope model (the CEM design). The average performance of models of the FEM design was consistently higher than those of the CEM design. There were significant differences in model performance among species but there was no significant interaction between model design and species. The average maximum kappa value ranged from 0.70 to 0.90 for FEM design and from 0.57 to 0.89 for the CEM design, which can be described as 'good' to 'excellent' using published ranges of agreement for the kappa statistic. This technique can be used to predict species' potential distributions that could be used for identifying regions at risk from invasion by alien species. These predictions could also be used in conservation planning in the case of native species.

One hundred years of botany at Rhodes University

An indigenous garden was created in its courtyard, mainly of species of the Eastern Cape province, and its growth in diversity and stature mirrors the changes in the department over the years. Professor Selmar Schonland, the founding father of botany at Rhodes, was also the curator/director of the Albany Museum. Under his leadership, systematics was firmly established as a discipline at Rhodes and research on the flora of the Eastern Cape begun, to be continued to this day. Under professors William Isaac (1949–1951) and Edgar Twyman (1951–1973), plant physiology became an important area of study, and future leaders in physiology at other universities obtained degrees under them. Phycology and aquatic ecology became notable disciplines with Mary Pocock’s world-class work in the 1940s on freshwater and marine algae, which inspired others such as head of department Professor Stanley Seagrief (1958–1986) to develop these studies further. Mycology and microbiology were established as major subdisciplines under Noel Smith (1926–1948) and Twyman through to the 1960s, when the Department of Microbiology was established. Early studies in ecology owe their success to Tony Martin (1947–1956) and the link with the Grahamstown Botanical Research Station, when a sound base for plant ecology in the Eastern Cape was established. Coastal ecology became a major focus in the 1970s, when Roy Lubke and Ted Avis concentrated on dunes and the management of coastal regions. This led to the establishment of the Department of Environmental Science in the 1990s. The expertise of the botany staff in teaching and research in a variety of fields is reflected in the careers of its many students around the world.

- Description: Chlorophyll fluorescence induction curves have been used as a sensitive tool for screening the photosynthetic performance of plants. Experimental treatments involving nitrate supply and chilling stress have been shown to affect fluorescence induction curves and other measures of photosynthesis. We have investigated the photosynthetic performance of Sorghum bicolor supplied with Long Ashton growth solution containing standard (20 μmol mol^(–1)) or low (5 μmol mol^(–1)) phosphorus. The JIP-test based on the chlorophyll fluorescence induction curve was used as a non-destructive method to measure the relative proportions of energy dissipated by different processes (termed energy fluxes) in the light reactions. The various energy fluxes or derived parameters were compared to find the measures that were most sensitive to the experimental conditions. Plant response to treatments was first evident in selected chlorophyll fluorescence parameters, particularly performance index (PI_ABS_); plants with increased PI_ABS_ manifested higher electron transport activity and dissipated less energy as heat, possibly as a result of their better phosphorus status, leading to more functional reaction centres. Observed changes in fluorescence were correlated to changes in gas exchange and biomass. Standard phosphorus treatments significantly increased biomass, leaf area, photosynthetic and respiratory rates, carboxylation efficiencies and levels of ribulose biphosphate regeneration rates, relative to plants with low supplies of nutrients.

The JIP-test based on the chlorophyll fluorescence induction curve was used as a non-destructive method to measure the relative proportions of energy dissipated by different processes (termed energy fluxes) in the light reactions. The various energy fluxes or derived parameters were compared to find the measures that were most sensitive to the experimental conditions. Plant response to treatments was first evident in selected chlorophyll fluorescence parameters, particularly performance index (PI_ABS_); plants with increased PI_ABS_ manifested higher electron transport activity and dissipated less energy as heat, possibly as a result of their better phosphorus status, leading to more functional reaction centres. Observed changes in fluorescence were correlated to changes in gas exchange and biomass. Standard phosphorus treatments significantly increased biomass, leaf area, photosynthetic and respiratory rates, carboxylation efficiencies and levels of ribulose biphosphate regeneration rates, relative to plants with low supplies of nutrients.

Molecular chaperones can be defined as proteins that facilitate the correct folding of other proteins, so that they attain a stable tertiary structure. In addition, they promote the refolding and degradation of denatured proteins after cellular stress. Heat shock proteins form one of the main classes of molecular chaperones. We are interested in determining if the genome of the coelacanth (Latimeria chalumnae) encodes a heat shock protein-based cytoprotection mechanism. We have isolated 50 kb and larger coelacanth genomic DNA from frozen skin tissue of L. chalumnae. From the alignments of several fish Hsp70 proteins, conserved regions at the N- and C-termini were identified. Codon usage tables were constructed from published coelacanth genes and degenerate primers were designed to isolate the full-length hsp70 gene and regions encoding the ATPase and the peptide binding domains. Since it is known that the tilapia and Fugu inducible hsp70 genes are intronless, we proceeded on the assumption that a coelacanth inducible hsp70 would also be intronless. A large fragment (1840 bp) encoding most of a coelacanth Hsp70 protein, and two partial fragments encoding a coelacanth Hsp70ATPase domain (1048 bp) and peptide binding domain (873 bp), were isolated by polymerase chain reaction amplification. Protein sequences translated from all the nucleotide sequences were closely identical to typical Hsp70s. This is the first study to provide evidence for a cytoprotection mechanism in the coelacanth involving an inducible Hsp70.

Root samples from 17 tree species common to the pockets of forest in the Eastern Cape province, and representing the families Rubiaceae, Scrophulariaceae, Oleaceae, Podocarpaceae, Myrsinaceae, Anacardiaceae, Caesalpinoideae, Papilionoideae, Rutaceae, Meliaceae, Celastraceae, Flacouticeae and Ebenaceae, were sampled and examined for mycorrhizal colonization. Microscopic examination of all the species produced evidence of morphological structures indicative of endomycorrhizal associations as indicated by the presence of intercellular hyphae combined with vesicles, arbuscules or hyphal coils. Hyphal coils (also known as Paris-type associations) appeared to be abundant, especially within the Cassine genus. Arbuscules (also known as Arum-type associations) were scarce but sometimes present, and vesicles were prolific in Olea capensis. - Description: The process of assisted protein folding, characteristic of members of the heat shock protein 70 (Hsp70) and heat shock protein 40 (Hsp40) molecular chaperone families, is important for maintaining the structural integrity of cellular protein machinery under normal and stressful conditions. Hsp70 and Hsp40 cooperate to bind non-native protein conformations in a process of adenosine triphosphate (ATP)-regulated assisted protein folding. We have analysed the molecular chaperone activity of the cytoplasmic inducible Hsp70 from Trypanosoma cruzi (TcHsp70) and its interactions with its potential partner Hsp40s (T. cruzi DnaJ protein 1 [Tcj1] and T. cruzi DnaJ protein 2 [Tcj2]). Histidine-tagged TcHsp70 (His-TcHsp70), Tcj1 (Tcj1-His) and Tcj2 (His-Tcj2) were over-produced in Escherichia coli and purified by nickel affinity chromatography. The in vitro basal specific ATP hydrolysis activity (ATPase activity) of His-TcHsp70 was determined as 40 nmol phosphate/min/mg protein, significantly higher than that reported for other Hsp70s. The basal specific ATPase activity was stimulated to a maximal level of 60 nmol phosphate/min/mg protein in the presence of His-Tcj2 and a model substrate, reduced carboxymethylated α-lactalbumin. In vivo complementation assays showed that Tcj2 was able to overcome the temperature sensitivity of the ydj1 mutant Saccharomyces cerevisiae strain JJ160, suggesting that Tcj2 may be functionally equivalent to the yeast Hsp40 homologue (yeast DnaJ protein 1, Ydj1). These data suggest that Tcj2 is involved in cytoprotection in a similar fashion to Ydj1, and that TcHsp70 and Tcj2 may interact in a nucleotide-regulated process of chaperone-assisted protein folding.

The natural products chemistry of southern Africa’s unique marine flora and fauna is relatively unknown and this review provides an overview of the contribution made by the marine natural products research group at Rhodes University to the isolation, identification and synthesis of biologically active natural products from southern African marine microorganisms, algae, sponges, ascidians, soft corals and molluscs.
