- Description: Porous CaCO3 microparticles have been used earlier for sustained drug release of hydrophilic drugs but have certain drawbacks for use with hydrophobic drugs. Hence, to overcome these drawbacks, a novel composite of CaCO3 along with cyclodextrin (CD–CaCO3) for the delivery of hydrophobic drugs was developed. Cyclodextrins (CDs), when incorporated within CaCO3, increased the porosity and surface area of microparticles thereby enhancing the encapsulation efficiency of hydrophobic drugs (5-Fluorouracil or Na-L-thyroxine) by forming inclusion complexes with cyclodextrin. Thermogravimetric and FTIR studies confirmed the interaction between the cyclodextrin and CaCO3 microparticles. Raman spectra confirmed the peak of vaterite crystals before and after loading of hydrophobic drugs within the composite. In vitro release studies when performed at pH 4.8 (5-Fu) and pH 1.2 (Na-L-thy) showed release at low pH as CaCO3 is soluble at acidic pH unlike slower release at basic pH. Release kinetics followed a Higuchi kinetic model at pH 4.8 (5-Fu) and pH 1.2 (Na-L-thy) respectively.

Analytical methods for the assessment of pharmaceutical product quality must be validated according to regulatory guidelines to ensure that tests are reliable and valid. Agitation rate, mesh pore size, surfactant concentration, and dissolution medium molarity are experimental parameters that may affect nevirapine (NVP) release and were investigated and optimized to ensure that consistent, reliable, and valid results using Apparatus 3 were produced. Agitation rate was investigated to establish an equivalent response to that observed for NVP release using Apparatus 2 at 50 rpm. A reciprocation rate of 5–10 dpm produced dissolution profiles that were similar to those observed using Apparatus 2. An increase in the molarity of the dissolution medium slightly increased the release rate of NVP, and a 50 mM buffer maintained at pH values mimicking gastrointestinal tract (GIT) conditions was selected for all experiments. With the addition of 2% sodium lauryl sulfate (SLS) to the dissolution medium, >80% NVP was released from the tablets over the test period. The NVP release rate increased with an increase in the mesh pore size; however, the extent of release was not affected by this parameter. Dissolution test samples were analyzed using HPLC, and dissolution methods were validated for NVP stability in the dissolution medium, specificity, linearity and range, repeatability, intermediate precision, and accuracy as defined by ICH. The dissolution method used for testing NVP tablets can be regarded as an appropriate tool for the evaluation of sustained-release (SR) NVP formulations and the impact of formulation composition and product quality attributes on drug release.

Stable isotope and fatty acid data both revealed spatial and developmental variations in the shrimp diet. Nearshore shrimp were more 13C-enriched than those from the inter-island region, suggesting increased kelp detritus entered the food web in the nearshore regions. The shrimp showed increases in δ13C and δ15N signatures (and trophic position) with an increase in body size, resulting in distinctions between size classes that reflected shifts in their trophic niche through development. The fatty acid profiles similarly indicated distinctions in diet with increased shrimp size (in the deep regions), and spatial variability was evident in relation to region and depth. All shrimp contained large proportions of polyunsaturated and essential fatty acids, indicating that the quality of food consumed was similar between regions despite the diet variability. Our results provide new dietary information about a key species operating near the base of the food web at the highly productive PEI, and show that there were no areas of enhanced nutrition available to the shrimp. As such, there was no nutritional advantage to shrimp inhabiting any specific region around the PEI.

Our results provide new dietary information about a key species operating near the base of the food web at the highly productive PEI, and show that there were no areas of enhanced nutrition available to the shrimp. As such, there was no nutritional advantage to shrimp inhabiting any specific region around the PEI.

The purpose of this review is to summarise the challenges and future prospects associated with biological water quality monitoring using diatoms with special focus on southern Africa. Much work still needs to be carried out on diatom tolerances, ecological preferences and ecophysiology. It is recommended that past research pertaining to African diatom taxonomy should be made readily accessible to all through electronic media for use as a reference point. Moreover, following the same approach as for macroinvertebrate biomonitoring, African and other developing countries can resort to intermediate diatom taxonomy (i.e. genus), which is easier, less time-consuming and requires less-skilled personnel. While the lack of capacity and baseline information on diatom community composition and ecological requirements represent significant hurdles, diatom biomonitoring potentially holds much promise for understanding the ecological functioning and management of aquatic ecosystems in southern Africa. The application of diatom-based water quality assessment protocols has direct and immediate value for use as an ‘added-value’ assessment tool in addition to the use of macroinvertebrates and fish indices as these can indicate anthropogenically impacted and pristine sites.

With the exception of Cr and Pb, concentrations of elements were higher than their background in soil, especially for Cd (1.79 mg kg−1), As (99.61 mg kg−1) and Ni (142.62 mg kg−1), which were 18.49, 8.89 and 5.30 times their background concentrations, respectively. The whole sampled zone was characterized by medium pollution and had a very high potential ecological risk. The area of arable land presented a medium pollution risk, while areas near to the path or road showed high risk of pollution. The predominant contributors to elevated ecological risk for the whole zone were Cd and As.

The species richness was highest in the middle reach, decreasing from early spring to summer in all reaches. The results of the study provide important insights into the various factors that structure benthic diatom community composition within a permanently open temperate southern African estuary.

Additionally, for birds, the proximity to green space, tree size and species were also important, whilst for mistletoes, the proximity to green space, slope aspect, and tree species were significant. Preference ratios indicated that some tree species had a higher than random occurrence of birds or mistletoes, whilst others had a low abundance. The indigenous tree species, Acacia karroo Hayne was the only reasonably abundant street tree species that was important for birds, nests, and mistletoes. At the street scale, there was a positive relationship between street tree species richness and bird species richness. These results emphasise the importance of selecting appropriate tree species if biodiversity conservation is a core outcome.

- Description: publisher version , This interaction of silver nanoparticles (AgNP) with human immune-deficiency virus aspartic protease (HIVPR) is examined by molecular dynamics simulation using the Colores (Situs) package and biophysical techniques using UV–vis spectroscopy, dynamic light scattering, transmission electron microscopy and circular dichroism. The ‘docking’ of AgNP with HIVPR creates a complex [AgNP–HIVPR] to initiate a hypochromic time-dependent red-shift for the surface plasmon resonance maximum. MD simulations reflect large perturbations to enzyme conformations by fluctuations of both rmsd and B-factors. Increase in changes to electrostatic potentials within the enzyme, especially, with chain B, suggest hydrophobic interactions for the binding of the AgNP. This is supported by changes to mainchain and sidechain dihedrals for many hydrophobic amino acid including Cys95, Trp6 and Trp42. Circular dichroism spectra reveal disappearance of α-helices and β-sheets and increase in random coil first from chain B then chain A. During initial stages of the interactive simulation the enzyme is conformational flexible to accommodate the AgNP, that docks with the enzyme under a cooperative mechanism, until a more stable structure is formed at convergence. There is a decrease in size of the HIVPR–AgNP complex measured by changes to the gyration radius supporting evidence that the AgNP associates, initially, with chain B.

- Description: publisher version , This interaction of silver nanoparticles (AgNP) with human immune-deficiency virus aspartic protease (HIVPR) is examined by molecular dynamics simulation using the Colores (Situs) package and biophysical techniques using UV–vis spectroscopy, dynamic light scattering, transmission electron microscopy and circular dichroism. The ‘docking’ of AgNP with HIVPR creates a complex [AgNP–HIVPR] to initiate a hypochromic time-dependent red-shift for the surface plasmon resonance maximum. MD simulations reflect large perturbations to enzyme conformations by fluctuations of both rmsd and B-factors. Increase in changes to electrostatic potentials within the enzyme, especially, with chain B, suggest hydrophobic interactions for the binding of the AgNP. This is supported by changes to mainchain and sidechain dihedrals for many hydrophobic amino acid including Cys95, Trp6 and Trp42. Circular dichroism spectra reveal disappearance of α-helices and β-sheets and increase in random coil first from chain B then chain A. During initial stages of the interactive simulation the enzyme is conformational flexible to accommodate the AgNP, that docks with the enzyme under a cooperative mechanism, until a more stable structure is formed at convergence. There is a decrease in size of the HIVPR–AgNP complex measured by changes to the gyration radius supporting evidence that the AgNP associates, initially, with chain B.

However, the cycling of carbon in the soil under applications of mulch on sloping arable land is not yet fully understood. A four-year field experiment was carried out in Xiaofuling watershed of Danjiangkou reservoir in China. The object was to evaluate the effects of the application of straw mulch (ST) and grass mulch (GT) on dynamic changes in soil organic carbon and its fractions. Results showed that mulch applied on the soil surface increased the contents of SOC and its active fractions in the soil. Compared to the control without cover (CK), ST and GT treatments increased the contents of SOC, LOC, DOC, POC and EOC by 14.73%, 16.5%, 22.5%, 41.5% and 21%, respectively, in the 0–40 cm soil layer, and by 17%, 14%, 19%, and 30%, respectively, in the 0–100 cm soil layer. The contents of organic carbon and its active fractions decreased with increasing soil depth in all of the treatments. SOC was accumulated in the period of December to the following March. The contents of soil DOC and LOC were high in January to March, while the contents of soil POC and EOC were high in June to September. The relative contents of soil organic carbon fractions were POC > EOC > LOC > DOC over the four years. Straw mulching had no significant effect on the changes in soil organic carbon active fractions during the different periods. Based on this long-term field experiment in Danjiangkou reservoir, we found that straw mulching had a significant effect on soil, increasing SOC content and stock in slopping arable land, and that live grass mulching was more effective than rice straw mulching. We discuss possible optimal periods for the implementation of mulching practices on sloping land.

In such cases, suboptimal management outcomes may, in addition to other factors, be symptomatic of a divergence in perspectives among these actors driven by fundamental differences in ecological knowledge. We examine the degree of congruence between the understandings of actors surrounding key issues of management concern in three case studies from tropical, subtropical and boreal forests. We identify commonly encountered points of divergence in ecological knowledge relating to key management processes and issues. We use these to formulate seven hypotheses about differences in the bodies of knowledge that frequently underlie communication and learning failures in forest management contexts where multiple actors are involved and outcomes are judged to be suboptimal. Finally, we present a set of propositions to acknowledge and narrow these differences. A more complete recognition of the full triangulation between all actors involved, and of the influence that fundamental differences in ecological knowledge can exert, may help lead to a more fruitful integration between local knowledge and practice, manager knowledge and practice, and contemporary science in forest management.
