Metabolic effects brought about by tricyclic antidepressants and the contribution of a medicinal plant in alleviating high fat diet induced insulin resistance in male wistar rats

The median age for diagnosis was around sixty, but recent surveys have shown that the entire age distribution curve shifting left. The incidence of type II diabetes is thought to be parallel with the growing rate of obesity associated with an unhealthy western diet. Type II diabetes is an expensive disease to manage, it is for this reason that cheaper medication needs to be investigated in the form of traditional plants, such as Sutherlandia frutescens. Prescription medication, such as tricyclic antidepressants, may also increase body weight thereby playing a role in obesity. The cause of weight gain in such cases may go unrecognized or lead to cessation of the medication with or without the practitioner’s knowledge or approval. It is therefore necessary to investigate the causative agents responsible for the excessive weight gain. Drinking water containing extracts of S. frutescens or metformin was administered to two groups of eleven insulin resistant male Wistar rats. The insulin resistant control group received water without any medication. Rats were sacrificed after 8 weeks allowing for fasting blood glucose, insulin and tissue glycogen content determination. Glucose uptake was also determined using [3H] deoxyglucose. The effect of the medication and the diet on muscle post receptor insulin signaling proteins was determined through Western blots. Liver proteomics was also performed using 2-D electrophoresis. In a separate experiment 26 male Wistar rats were exposed to strepotozotocin toxin, 7 of these rats received intravenous insulin treatment, 7 rats received S. frutescens extract and 7 rats received a combination of both medications, the remaining 5 received no treatment and served as the control. Rats were sacrificed after 6 days allowing for fasting blood glucose, insulin and tissue glycogen content determination. Two groups of 14 male Wistar rats received amitriptyline or trimipramine (common tricyclic antidepressants) in their drinking water, the control group (30 rats) received water without any medication. The rats’ weight and food consumption was monitored throughout the trial and their oxygen consumption was also determined. Rats were sacrificed after 6 weeks or 14 weeks of medicinal compliance allowing for fasting blood glucose, insulin and tissue glycogen content determination. Glucose uptake was also determined using [3H] deoxyglucose. S. frutescens treatment normalized circulating serum insulin levels and significantly increased the rate of glucose clearance. Certain post receptor insulin signaling proteins were also significantly increased relative to the insulin resistant control group. 2-D electrophoresis identified the normalization of protein levels associated with the urea cycle. S. frutescens was also able to, independently; maintain normoglycaemic levels in the strepotozotocin treated group. The tricyclic antidepressants significantly increased blood glucose levels while significantly reducing tissue glycogen levels for both sacrifice periods. Serum insulin remained unchanged while a significant increase in insulin degradation and insulin degrading enzyme levels were found for both antidepressants. S. frutescens shows promise as a low cost antidiabetic medication for future use. Although the antidepressants did not promote weight gain, the increase in blood glucose levels may be cause for concern in patients with a pre-disposition toward developing diabetes.

Targeted contractors are those who do not have the know-how necessary to develop such a WSP. Thus the need for a basic approach or model that will, through its simplicity and ease of application, assist and encourage such contractors nonetheless to undertake the process. Achieving this objective required a detailed examination of relevant literature and legislation, to identify the basic or minimum requirements for developing a strategic WSP and for meeting grant recovery regulations respectively. Newly released grant recovery regulations were examined to identify the implications thereof, and to establish the minimum legislative requirements, for the WSPs of small and micro contractors. Existing relevant general training and training needs assessment models were examined to identify the requirements for developing a strategic WSP. To establish the minimum strategic requirements it was necessary to firstly identify all the requirements proposed by the different models for identifying and meeting the training needs of an organisation, and thereafter reach a decision regarding which could be considered as minimum requirements. The decision rule for selection as a minimum strategic requirement was inclusion in all the surveyed models. To enable a less subjective analysis than relying solely on the opinion of the researcher, a content analysis was selected as the research technique, as the steps of a typical content analysis include a number of measures to increase objectivity. The legislative and strategic requirements identified by the study were converted into a series of simple sequential action steps to formulate a practical model that would guide the targeted contractors through the process of developing a value-adding WSP.

The effect of different slaughter age regimes on the primary and secondary production parameters of Ostriches (Struthio Camelus Domesticus) and the economic consequences of different slaughter ages

- Description: The South African ostrich industry is presently characterised by a drastic decline in the slaughter price of slaughter birds, due to the drop in the price of meat as well as leather. As a result, the economic viability of ostrich farming is severely hampered by these developments, and many generally accepted industry practices have to be reviewed. Among the practices under review is the possibility of slaughtering ostriches at an earlier age than the traditional slaughter age of 12 - 14 months, in order to minimise feed costs and the incidence of damage to the skin. However, little information is available on the effect of the slaughter age on the yield and quality of the economically important end-products. The effect of slaughter age on production and the economic viability of a commercially intensive ostrich production system was consequently evaluated in a completely randomised experimental design involving two batches (n= 81 and n= 94) of ostriches. The experiments took place at Kromme Rhee experimental farm of the Elsenburg Research Centre near Stellenbosch. The birds were subject to experimental treatment at the age of 4 months (n= 81, from May 2004 to March 2005) and 6 months (n= 94, from August 2004 to August 2005) respectively. The ostriches were randomly divided into 10 groups of between 16 and 20 birds per group. Standard production practices, as applied in an intensive ostrich production unit, were implemented. Clean drinking water and self-mixed grower diet (min 15 percent crude protein, 0.68 percent lysine and 11.4 MJ ME per kg feed) and finisher diet (min 14 percent crude protein, 0.55 percent lysine and 9.8 MJ ME per kg feed) were provided ad libitum to the birds. Birds were slaughtered at the ages of 8.5, 10.5, 12.5, 14.5, and 16.5 months respectively. Data on feed intake and yields of cold carcass, crust skin size, skin grade and total feathers was recorded for birds slaughtered at the respective ages. Data was analysed according to the analysis of variances. Values for meat yield, skin size yield and feed intake were predicted by regression analysis based on experimental values. Industry figures and norms were used as proxy for other production inputs and costs. A gross margin type analysis was performed to evaluate the effect of different slaughter ages on the profitability of each production system. There was a positive correlation between age and feed intake (r2= 0.40), slaughter weight (r2= 0.97), cold carcass yield (r2= 0.48), skin surface area (r2= 0.39), skin grade (r2= 0.19) and total yield of feathers (r2= 0.29). The dry skin grade showed deterioration as slaughter age was prolonged. Highly significant differences (P< 0.01) in slaughter weight, cold carcass yield, skin surface area, dry skin grade and total feathers were detected at the different slaughter ages. There were no significant differences (P> 0.05) with respect to gender for slaughter weight, skin surface area and dry skin grade within the different age groups. Males had significantly higher (P< 0.05) cold carcass yield and total feather yield than their female counterparts. From this study it can be concluded that an increase in slaughter age significantly increases feed intake, slaughter weight, cold carcass yield, and skin surface area. It is also clear that an increase in slaughter age can be detrimental to dry skin grade. The relative income contribution of the three main products (skin: meat: feathers) occurred in a ratio of 47:53:0, 52:47:1, 47:50:3, 44:51:5, 39:56:5 at the respective slaughter ages. Total income revealed an increase of 26 percent from 8.5 to 10.5 months slaughtering, an increase of 4 percent between 10.5 and 12.5 months, and an increase of 11 percent between 12.5 and 14.5 months. The income dropped by 0.3 percent between 14.5 and 16.5 months slaughtering. Total costs increased with slaughter age, particularly feed costs which showed an increase (at a decreasing rate) between the different slaughter ages. The margin above specified cost per bird was the highest for birds slaughtered at the age of 10.5 months. The margin above cost showed a steady decline as birds were slaughtered at higher ages. Slaughtering at 16.5 months revealed a negative margin. To compare the effect of differences in time span on different slaughter age regimes, margin above costs for each system over 5 years were discounted to a present value. In this study, with current feed costs and income from end products, slaughtering at 10.5 months revealed the highest present value for the margin above cost. Furthermore, slaughtering at 16.5 months revealed a negative present value on the margin above cost after 5 years. Financial viability calculations are relevant and valid for data obtained in this case study. Financial viability is however case-specific and may vary between producers, depending on their individual management practices, production systems and cost structures. Results from this study may be of significant value to managerial decision-making in the ostrich industry.

An investigation into the neuroprotective properties of the non-steroidal anti-inflammatory agents tolmetin, sulindac and turmeric

- Description: Accumulating evidence suggests that anti-inflammatory agents and antioxidants have neuroprotective properties and may be beneficial in the treatment of neurodegenerative disorders. In the present study, the possible neuroprotective properties of tolmetin, sulindac and turmeric were investigated. The antioxidant effects of tolmetin and sulindac were determined by inducing free radical generation with quinolinic acid (QA), cyanide or iron (II) in rat brain homogenates or primary hippocampal neurons. Tolmetin and sulindac significantly reduce lipid peroxidation and scavenge the superoxide anion. Metal binding studies were conducted to determine whether metal chelation is a possible mechanism through which these agents reduce QA and iron (II)-induced lipid peroxidation. UV/VIS, infrared spectroscopy as well as electrochemical studies show that both agents bind to iron (II) and/or iron (III). Histological examination of the hippocampus showed that pre-treatment of animals with tolmetin or sulindac offers protection against intrahippocampal injections of QA. These agents also attenuate QA-induced apoptosis and reduce the loss of neurons in the hippocampus. The co-incubation of primary hippocampal neurons with the NSAIDS also enhanced cell viability which is significantly reduced by QA. Behavioural studies using a water maze showed that the treatment of animals after QA-induced neurotoxicity reduces QA-induced spatial memory loss. Tolmetin and sulindac also reduced glutathione depletion and protein oxidation in rat hippocampus. Both NSAIDS inhibit liver tryptophan 2,3-dioxygenase activity in vitro and in vivo and subsequently increased hippocampal serotonin levels. However, both NSAIDS also reduce dopamine levels in rat striatum. Tolmetin but not sulindac increased the synthesis of melatonin by the pineal gland. The active components of turmeric known as the curcuminoids were separated using preparative thin layer chromatography (TLC). The purity was confirmed by TLC, NMR and mass spectrometry. The environmental toxin lead, induces lipid peroxidation and reduces primary hippocampal neuronal viability. The co-incubation of the neurons with the curcuminoids significantly reduces lead-induced lipid peroxidation and enhances neuronal cell viability in the presence of lead. Lead-induced spatial memory deficit is also attenuated with curcumin, demethoxycurcumin but not bisdemethoxycurcumin. The curcuminoids also reduce lead-induced hippocampal glutathione depletion and protein oxidation. Metal binding studies show that the curcuminoids bind to lead and is another possible mechanism through which the curcuminoids reduce lead-induced neurotoxicity. The findings of this study indicate a possible role of tolmetin, sulindac and turmeric in neurodegenerative disorders such as Alzheimer’s disease. However, tolmetin and sulindac reduce dopamine levels.

UV/VIS, infrared spectroscopy as well as electrochemical studies show that both agents bind to iron (II) and/or iron (III). Histological examination of the hippocampus showed that pre-treatment of animals with tolmetin or sulindac offers protection against intrahippocampal injections of QA. These agents also attenuate QA-induced apoptosis and reduce the loss of neurons in the hippocampus. The co-incubation of primary hippocampal neurons with the NSAIDS also enhanced cell viability which is significantly reduced by QA. Behavioural studies using a water maze showed that the treatment of animals after QA-induced neurotoxicity reduces QA-induced spatial memory loss. Tolmetin and sulindac also reduced glutathione depletion and protein oxidation in rat hippocampus. Both NSAIDS inhibit liver tryptophan 2,3-dioxygenase activity in vitro and in vivo and subsequently increased hippocampal serotonin levels. However, both NSAIDS also reduce dopamine levels in rat striatum. Tolmetin but not sulindac increased the synthesis of melatonin by the pineal gland. The active components of turmeric known as the curcuminoids were separated using preparative thin layer chromatography (TLC). The purity was confirmed by TLC, NMR and mass spectrometry. The environmental toxin lead, induces lipid peroxidation and reduces primary hippocampal neuronal viability. The co-incubation of the neurons with the curcuminoids significantly reduces lead-induced lipid peroxidation and enhances neuronal cell viability in the presence of lead. Lead-induced spatial memory deficit is also attenuated with curcumin, demethoxycurcumin but not bisdemethoxycurcumin. The curcuminoids also reduce lead-induced hippocampal glutathione depletion and protein oxidation. Metal binding studies show that the curcuminoids bind to lead and is another possible mechanism through which the curcuminoids reduce lead-induced neurotoxicity. The findings of this study indicate a possible role of tolmetin, sulindac and turmeric in neurodegenerative disorders such as Alzheimer’s disease. However, tolmetin and sulindac reduce dopamine levels.

Data regarding the uncertainties from educators have been obtained from the SOC (Stages of Concern) questionnaires of Hall, George and Rutherford. The rest of the questionnaire concerned the biographical information regarding the educators as well as the support that educators have received during the implementation of the learning area. It emerged from the questionnaire that educators understand the importance of a new Natural Science curriculum for SA. They are the customary methods of presenting the elements of the text. There are no strict rules followed in the writing of plays, but there are conventions which vary from one playwright to another and from time to time. Conventions are the devices and the features of a literary work by which its kind can be recognized. Change creates anxiety, uncertainty and stress. Adaptation of culture to some changes plays a role as time passes by. To adapt to change is to be able to manage change. Managing change demands three levels of human response namely: the individual, the group and the cultural or social context. No matter how many changes are brought, different cultures should survive the changes for the nation to remain with its nationality. Industrialisation, urbanisation, religion, politics and economy are the agents of change. Also the social environment, human intelligence and culture play to a greater extent a role in the evolution process. Among other things, this study portrays that the changing times are reflected in Xhosa plays. This is the reflection of how people live, behave or do things, and think as time comes and passes. Pattern of development is traced through time, with the history being involved in the development. Change and development are unavoidable products of human thought. Development is traced from the primitive to the modern way of doing things. A modern or developed society is viewed as being capable of handling a wide variety of internal as well as external pressures. Every time a society manages a new pressure, its modernity improves. Thus, the word ‘modern’ has no time frame, as long as there is a new development, this term ‘modern’ features in. Though the study employs Evolutionary, Structuralist, Stylistic, Formalism and Marxist approaches, the branch of the Semiotic approach, Pragmatism, plays the major role in that the meaning of the texts is one of the semiotic categories. Again Semiotics deals with the writing and the interpretation of the text. Thus communication, adaptation and relating are fundamental to human existence and survival. It is easy to notice that there are old conventions that are continuing in the writings of the new generations of playwrights. This study compares and contrasts the similar conventions of dramatic texts, especially those that have the same theme and meaning. This study shows how the existing dramatic conventions are affected by time, history, economy, education, technology and some other changes. Though the dramatic conventions are said to be continuing, they also adapt to the changing time. There are conventional and cultural aspects that seem to be continuing, but it is a ‘changing continuity’. The developments or changes discussed in this study are in Xhosa drama conventions, those of culture of amaXhosa, dramatic construction of the Xhosa plays and in the interpretation of the plays.

Brine shrimp occur as either bisexual species or as parthenogenetic populations. Despite published reviews of their world-wide distribution little is known about their occurrence in Africa. This review adds new information about 70 African Artemia sites and lists 26 potential sites and their coordinates. Sixteen sites in Southern Africa and Namibia were visited during a collecting trip, and new information on the reproductive mode of nine of these sites is given. Several South African populations exhibit bisexual reproduction. In Namibia there are two parthenogenetic populations (Walvis Bay and Swartkops) and an additional bisexual population (Hentie's Bay). A mixed population (bisexual and parthenogenetic reproduction at the same site) was found at Coega, South Africa.

The lack of emotional intelligence, however, undermines both an individual’s and a company’s growth and success. Expressing emotions within the workplace has for many years been considered inappropriate and irrelevant to business. This research study points to the fact that an overwhelming amount of research shows that not only are emotions very much a part if the work experience but, to a large degree, set the course that companies should follow. This research study firstly discusses the core components of emotional intelligence and its impact on individuals and how they relate to others. The study further focuses on the impact that emotional intelligence has on aspects that contribute to organisational effectiveness. This study suggests that emotional intelligence within companies could result in a situation where employees communicate with understanding and respect and where confidence and enthusiasm in the organisation are instilled. Knowing one’s own emotions and controlling them; recognizing emotions in others and managing them; and self-motivation are key dimensions of emotional intelligence covered in the theoretical perspective of emotional intelligence discussed in Chapter Two. This study points to the benefits of emotional intelligence as an indispensable element not only in creating a profitable business but also in leading a successful life. The findings discussed in Chapter Six indicate a reasonable level of emotional intelligence amongst employees at GMSA who participated in the survey. Recommendations from the study indicated a need for training in various aspects of emotional intelligence in order to derive the benefits of improved organisational effectiveness and profitability.

Learning about approaches that might help organisations to develop, concepts such as organisation development in general and Future Search in particular, I developed an interest especially in the Future Search Conference. My interest was based on successful developmental stories of Future Search Conferences in organisations across the world. I decided to introduce the Future Search Conference to our organisation, which I thought might be a remedy for our problems at a later stage. The aim of the study was to investigate participants' experiences and the perception of the Future Search Conference as an approach to organisational change, through interviews. The study found that Future Search was a new concept/approach to the NHTN as well as to the whole Ministry, and it is different from the traditional strategic planning. The difference is seen in the way Future Search involves all stakeholders in issues of concern, while strategic planning involves only managers. The responses revealed the willingness of participants to implement the Future Search Conference in their organisation. Data also revealed, however, that it was not clear how ready the managers were to carry out this task, as they are the gatekeepers. Participants suggested a separate Future Search Conference with managers, to sensitise them to the approach, so that they can see whether it addresses the values and goals of the organisation and can be implemented for developmental purposes.

For this potential to be realised, the various educational resources that are commonly used by teachers and learners in rural schools need to incorporate natural resource management knowledge. Using Bernstein’s concepts of classification and his theories on recontextualisation, a content analysis study was carried out to compare the level of NRM integration within the Grade 10 Life Sciences syllabus, and a Grade 10 Life Sciences textbook. Results from the analysis of the syllabus showed that overall only 9% of all the knowledge statements analysed had a strong link to NRM and related issues. The highest percentage of such sentences was found in the Core Knowledge section of the syllabus (21%). For the textbook, only 8% of the analysed items had a strong link to NRM and related issues, with the highest percentage of such items occurring in the Suggested Activities section (16%). However, the level of NRM integration in both documents increased considerably when sentences that had only an implicit link to NRM and related issues were included. It was concluded that both documents provide ample opportunities for NRM learning, although the extent to which this occurs varies among their different sections. The recontextualising role of the Grade 10 Life Sciences textbook was reflected in its relatively higher level of NRM integration in the Suggested Activities category, and in the Glossary category. This study highlights the need for further strengthening of the position of NRM within the Grade 10 Life Sciences syllabus, and for more Bernstein-based research to inform South Africa’s curriculum reform initiatives in environmental education.

'Show and tell': a discursive analysis of women's written accounts of their self-injuring practices

- Description: Self-injuring is a practice that involves self-administered damage to one’s body, most commonly cutting of the skin on the forearms. (The practice is distinguished from other intentional and in/direct self-harmful or self-damaging behaviours that cause bodily harm). Dominant psychiatric, psychological or medical approaches construct self-injuring as deviant, socially unacceptable or abnormal behaviour that is indicative of more or less severe psychopathology, and importantly as a stereotypically female practice. This research is conducted within a post-essentialist framework and views self-injuring, and the injured body, as discursively constituted as well as a cultural and political act. It therefore moves away from pathologising discourses in which those who self-injure typically find themselves and their own accounts of their behaviour invalidated and silenced. Instead, the mental health perspective is viewed as one party among many that may contribute to the conceptualisation of ‘self-injuring’ practices as socially meaningful and thus self-injuring is critically interpreted without reliance on a medical model of ‘normalcy’. As part of attempts to challenge medical models and cultural ideals of normalcy, this research presents a critical discursive analysis of a series of narratives provided by 5 female participants in which they record their own experiences, feelings and thoughts related to their practices of selfinjuring. It makes use of critical discourse analytic methodology to identify certain characteristics of these narratives as representations of larger collective meaning systems. It analyses the ways in which self-injuring is constructed in women’s stories of their self-injuring experiences, focusing particularly on the subject positions available in these discourses, as well as their ideological effects. The analysis focuses particularly on constructions of the body and subject positions as they enable or undermines the self-injuring subject’s agency. Finally, it attempts to determine the limitations of certain accounts of self-injuring, pursuing multiple meanings of self-injuring and illuminating new dimensions of talk on self-injuring and novel ways of conceptualising and understanding the practice.

This noticeable deficiency in the literature marked the start of the current longitudinal study which was designed to provide information on the career development of nine to twelve-year old South African children. In South Africa research focusing on preadolescent career development is limited with existing research indicating the need to further explore how children’s career choice process initially develops. The study aimed to explore and describe the changes that occur, if any, over a four-year period in terms of occupational interest typology, occupational status levels, occupational gender stereotypes and occupational gender traditionality. Both developmental and career developmental theories were used to provide a context for exploring this process of preadolescent career development. It was decided that quantitative research methods would provide the necessary structure needed to conduct the research. Although semistructured interviews and biographical questionnaires were used to record the verbal data, the data obtained was transposed into nominal data for analysis. Participants responded to questions within a semi-structured interview that focused on four broad areas of questioning. These questions explored participants’ occupational aspirations, how many occupations they knew about, how much they knew about the identified occupation, and the extent to which they held gender-role stereotypes about certain occupations. Although much of the change has been to correct imbalances, inequities and disparities within our local communities as a result of Apartheid, change has also been motivated by National Government’s realisation that, as with governments throughout the world, there is a need to modernise all spheres of Government. Part of this transformation process at a local government level in South Africa has been to ensure that municipalities become more responsive to the communities’ needs. The guiding principles for this transformation are contained in the White Paper on the Transformation of the Public Service (1995) and the Batho Pele White Paper (1997). This has informed the Municipal Systems Act: Act 32 of 2000 of which Chapter 6 determines that municipalities will have a performance management system to promote a culture of performance management amongst the political structures, political office bearers, councillors and administration. The performance management system must ensure that the municipality administers its affairs in an economical, effective, efficient and accountable manner. A literature review contained in this research, indicates that internationally, implementing performance management systems at a local government level is impact upon by a number of factors such as the organizational culture of an institution. This research, which has been grounded within a constructivist paradigm, describes the impact organizational culture has had on the implementation process of the performance management system at Buffalo City Municipality. Interviews were conducted amongst the Section 57 employees (i.e. the Directors) and those employees directly responsible for implementing performance management. The four Directors, two General Managers and the portfolio councilor were interviewed. Semi-structured interviews were conducted with the interviewees and this culminated in a total of 10 hours of interviewing. The protocols were analyzed using the guidelines suggested by Boyatzis (1998) and the findings are detailed in two chapters. The findings of the research were that the implementation of the performance management system at Buffalo City Municipality has been impacted on by the dichotomy between the political and administrative leadership, resulting in non-implementation of council resolutions, the lack of an organizational strategy, poor institutional arrangements and inadequate resource allocation, are reported. However, the most important finding was the impact that a culture of fear has had on the implementation process. This culture of fear and its impact on the implementation of performance management system is described as the most pervasive and insidious of all the findings to have negatively impacted on the implementation process. This research ends with recommendations for further research and it is argued that each organization has its own unique organizational culture. The conclusion is that no single typology, as contained in the literature, which can account for the specific impact organizational culture will have on the implementation process of a performance management system at local government level in South Africa. Consequently, implementers of performance management systems must assess the unique characteristics of each organization’s culture prior to implementation, in order to evaluate its impact that the organizational culture can have on the process.

The findings of the research were that the implementation of the performance management system at Buffalo City Municipality has been impacted on by the dichotomy between the political and administrative leadership, resulting in non-implementation of council resolutions, the lack of an organizational strategy, poor institutional arrangements and inadequate resource allocation, are reported. However, the most important finding was the impact that a culture of fear has had on the implementation process. This culture of fear and its impact on the implementation of performance management system is described as the most pervasive and insidious of all the findings to have negatively impacted on the implementation process. This research ends with recommendations for further research and it is argued that each organization has its own unique organizational culture. The conclusion is that no single typology, as contained in the literature, which can account for the specific impact organizational culture will have on the implementation process of a performance management system at local government level in South Africa. Consequently, implementers of performance management systems must assess the unique characteristics of each organization’s culture prior to implementation, in order to evaluate its impact that the organizational culture can have on the process.
