- Description: The Russian Wheat Aphid (Diuraphis noxia, RWA) negatively impacts commercially grown barley and wheat in South Africa. Climate change, the attendant rise in [CO2], and the appearance of new RWA biotypes have the potential to induce severe crop yield loss in agriculturally important wheat and barley cultivars. This study presents data showing changes in relative aphid population numbers, concurrently with assessments of plant damage under controlled environmental conditions, under ambient and elevated (450 ppm) [CO2]. Extensive structural damage to the vascular tissue and disruption to the transport systems were revealed using light, fluorescence and electron microscopy. This, coupled with biotype population studies, demonstrated that RWA has the capacity to inflict severe, potentially permanent damage to vegetative small grain plants. Furthermore, some currently ‘resistant’ cultivars may well lose resistance as a direct result of increasing atmospheric [CO2]. A small (50 ppm) increase in atmospheric [CO2] may result in increased aphid population numbers, potentially serious plant damage and, by implication, a potentially negative impact on yield, as increased aphid density per plant leads to an accelerated disruption of the assimilate and transpiration transport pathways. These outcomes pose a direct threat to the commercial small grain industry of South Africa and by extension, to other small grain production areas elsewhere.

Sea turtles as ocean ambassadors: opportunities and challenges

- Description: Sea turtles are ocean migrants that nest on the same beaches where they were born but forage on reefs and oceanic waters great distances away. Movement between these locations is sometimes years or even decades apart. Because of these broad-ranging movements and the many countries, they visit throughout their lives, effective conservation can only be achieved through international cooperation. However, wherever and whenever sea turtles come ashore, they fascinate people. Watching a sea turtle nest is like looking back through a window into deep time. This appearance and disappearing act of sea turtles create an enigma that elicits a multitude of disciplinary, inter-, and intradisciplinary teaching, research and engagement opportunities ranging from archaeology to social sciences, including tourism, biology and ecology, conservation and policy. In these different spheres, I operated over the last two decades to understand sea turtles, their biology and behaviour to affect their conservation. The biggest question I have pursued in my research career is to understand why the leatherback sea turtle population (Dermochelys coriacea) nesting in the iSimangaliso Wetland Park, South Africa, has not increased despite decades of protection. Another sea turtle species, namely loggerheads (Caretta caretta) nesting in the same area, experiencing similar conditions, has responded positively to conservation. Through two decades of research evaluating the intrinsic and extrinsic population drivers, such as reproductive output, age to maturity, natality and mortality, it seems evident that the population dynamics of sea turtles is much more complicated than what a simple population model would predict. From the literature, it is clear that other species, like the Mediterranean monk seal, red knot (a sandpiper) and other coastal species, are suffering a similar fate, i.e., lack of recovery despite conservation. These trends suggest that these species have become refugees in their own habitat. Marine habitats are transformed through human activities and may now be unsuitable to support larger populations under the current climate for these complex species. Current research is aimed to disentangle past and present distributions to assess if these species have responded by using alternative habitats over time or if there are body condition parameters (such as individual size, offspring size or survivorship, or metabolomics) that will point us in the direction to grow these endangered populations. Our research suggests that sea turtles, with their very complex life history facing multiple threats, live at the edge of success and extinction. Understanding and managing their path to success is a delicate balance with many aspects that need consideration.

In my presentation, I have mindfully taken cognisance of the plight of the so called lost generation, especially among our youth, those who have become strangers in their own land – no language, only misguided and distorted cultural values, no self, distorted history - this of course because of indoctrinations of some psycho-socio-histori-cultural and political imperatives. Strangely enough these are some of the issues that SEK Mqhayi warns us about in his creative works and has made efforts for us to see, but unfortunately no one listened or saw anything. Now that we “have the truth but denied the truth and now that we have the light, but we sit in darkness, Shivering, benighted in the bright noon-day sun,” and now that we are all blind, I think, this evening is the right time to see.

- Description: Molecular chaperones and their associated co‐chaperones play an important role in preserving and regulating the active conformational state of cellular proteins. The chaperone complement of the Indonesian Coelacanth, Latimeria menadoensis, was elucidated using transcriptomic sequences. Heat shock protein 90 (Hsp90) and heat shock protein 40 (Hsp40) chaperones, and associated cochaperones were focused on, and homologous human sequences were used to search the sequence databases. Coelacanth homologs of the cytosolic, mitochondrial and endoplasmic reticulum (ER) homologs of human Hsp90 were identified, as well as all of the major co‐chaperones of the cytosolic isoform. Most of the human Hsp40s were found to have coelacanth homologs, and the data suggested that all of the chaperone machinery for protein folding at the ribosome, protein translocation to cellular compartments such as the ER and protein degradation were conserved. Some interesting similarities and differences were identified when interrogating human, mouse, and zebrafish homologs. For example, DnaJB13 is predicted to be a non‐functional Hsp40 in humans, mouse, and zebrafish due to a corrupted histidine‐proline‐aspartic acid (HPD) motif, while the coelacanth homolog has an intact HPD. These and other comparisons enabled important functional and evolutionary questions to be posed for future experimental studies.

South Africa's "Rainbow People", national pride and happiness

- Description: Since the first free elections were held in April 1994, South Africans are popularly known as the ''rainbow people''. The paper inquires whether South Africans who experienced pride in their nation in the first years of democracy also perceived a greater sense of subjective well-being. It is proposed that national pride in post-apartheid South Africa might be fused with or work through self-esteem to lift levels of happiness. The paper traces the history of the new integrating civil religion of the rainbow people and the acceptance of the rainbow as a political symbol of unity among the diverse people of South Africa immediately after the 1994 elections and two years later. The proposed link between national pride and happiness was explored with data from two independent national surveys, the 1995 South African World Values Survey conducted by Markinor and a June 1996 MarkData syndicated omnibus survey. The study found that the appeal of the rainbow as political symbol was inclusive of all groups in society and that feelings of national pride and support for the rainbow ideal were positively associated with subjective well-being. As indicated by intensity and frequency measures, the majority of South Africans were proud of their country and could name a national achievement that inspired pride. Better-off South Africans tended to be happier and more satisfied with life but less proud, while the poor were less happy but fiercely proud of their country. Results suggest that belief in South Africa’s ''rainbow nation'' ideal may have assisted in boosting happiness during the transition to a stable democracy, thereby preventing alienation among the losers under the new political dispensation. Supporters of the ideal of the rainbow nation were more optimistic than others about the future of their country.

Gonadosomatic indices (GSI) varied between year-round consistency and apparent seasonal peaks in both summer and winter; there was no clear spatial pattern. Monthly percentage maturity provided further evidence for two peak reproductive periods each year, although mature squid were present throughout. Sex ratios demonstrated great variability between different areas and life history stages. Male-biased sex ratios were only apparent on the inshore spawning grounds and ranged between 1.118:1 and 4.267:1. Size at sexual maturity was also seasonal, squid maturing smaller in winter/spring than in summer/autumn. Also, squid in the east matured smaller than squid in the west. Although the results from the present study do not provide conclusive evidence of distinct geographic populations, squid likely spawn over a significantly larger area of the Agulhas Bank than previously estimated, and squid on the west coast of South Africa may return to spawn on the western portion of the Agulhas Bank. It remains likely, however, that the east and west coast populations are a single stock and that migration of juveniles to the west coast and their subsequent return as sub-adults is an integral but non-essential and variable part of the life history.

However, the mechanism of interaction of CA-VIII to ITPR1 is not well understood. In addition, functional defects to CA-VIII due to non-synonymous single nucleotide polymorphisms (nsSNVs) result in Ca2+ dysregulation and the development of the phenotypes such as cerebellar ataxia, mental retardation and disequilibrium syndrome 3 (CAMRQ3). - Description: The work reports on cyclic voltammetry (CV), square wave voltammetry and spectroelectrochemistry of the following complexes: tetrakis (benzyl mercapto) phthalocyanine complexes of Zn(II) (ZnTBMPc, 4a ), Co(II) (CoTBMPc, 5a ), Fe(II) (FeTBMPc 6a ); and tetrakis (dodecylmercapto) phthalocyanine complexes of Zn(II) (ZnTDMPc, 4b), Co(II) (CoTDMPc, 5b) and Fe(II) (FeTDMPc, 6b). - Description: The work reports on cyclic voltammetry (CV), square wave voltammetry and spectroelectrochemistry of the following complexes: tetrakis (benzyl mercapto) phthalocyanine complexes of Zn(II) (ZnTBMPc, 4a ), Co(II) (CoTBMPc, 5a ), Fe(II) (FeTBMPc 6a ); and tetrakis (dodecylmercapto) phthalocyanine complexes of Zn(II) (ZnTDMPc, 4b), Co(II) (CoTDMPc, 5b) and Fe(II) (FeTDMPc, 6b). More reversible CV couples were observed for complexes 4a, 5a and 6a containing thiol phenyl ring substituents. Complexes 4b, 5b and 6b containing long chain thiol substituents showed less reversible couples. Complexes 6a and 6b showed a relatively large number of redox processes (5 for 6a and 6 for 6b) within the potential window employed in this work. The Au-ME-CoTCAPc SAM sensor showed a very fast amperometric response time of approximately 1 s, good linearity at the studied concentration range of up to 5 μM with a coefficient R² = 0.993 and a detection limit of 0.4 μM oxidatively. Also reductively, the sensor exhibited a very fast amperometric response time (~1 s), linearity up to 5 μM with a coefficient R² = 0.986 and a detection limit of 0.2 μM. The cobalt tetracarboxylic acid phthalocyanine self-assembled monolayer was then evaluated as a mediator for glucose oxidase (GOx)-based biosensor. The GOx (enzyme) was immobilized covalently onto Au-ME-CoTCAPc SAM using coupling agents: N-ethyl-N(3-dimethylaminopropyl) carbodiimide (EDC) and N-hydroxy succinimide (NHS), and the results demonstrated a good catalytic behavior. Kinetic parameters associated with the enzymatic and mediator reactions were estimated using electrochemical versions of Lineweaver–Burk and Hanes equation, and the stability of the sensor was tested. The biosensor (Au-ME-CoTCAPc-GOx SAM) electrode showed good sensitivity (7.5 nA/mM) with a good detection limit of 8.4 μM at 3σ, smaller Michaelis–Menten constant (4.8 mM from Hanes plot) and very fast response time of approximately 5 s.

The Benthic invertebrate community of a Southern Cape estuary : structure and possible food sources

Infaunal bivalves comprised >60 % of the invertebrate biomass at Zostera sites but <5 % at bare sand sites. Conversely the infaunal anomuran C. kraussi dominated the sandy sites (>80 %) and was a minor component (<5 %) at eelgrass sites. The supratidal invertebrate community was dominated by Orchestia spp. which live and feed on wrack detritus. Litterbag experiments revealed that degradation of Zostera leaf wrack was rapid during the first thirty days after deposition but slow between 40 and 140 days. Laboratory experiments indicated that Orchestia consumption of wrack material could not account for the rapid weight loss recorded in the natural environment. Preliminary diet analyses of intertidal and infratidal zoobenthos revealed that most invertebrate species feed on detritus and associated microorganisms. Filamentous algae and diatoms dominated the gut contents of only three out of 18 macrobenthic species, and living Zostera was not an important food item for any invertebrate examined.

Recent demographic changes in formerly White universities are having a significant effect on the nature of interaction in formal discussion groups. Despite a common perception that at a tertiary level tutors and students are equally entitled to speak and all contributions equally valued, this article reveals that postgraduate seminars are sites of competition for the floor and that there are significant imbalances in participation by different groups in this competitive speaking environment; it is further argued that these imbalances reflect different (culture- and gender-specific) assumptions about what constitutes appropriate participation and also, to some degree, previous learned discourse patterns associated with schooling experience.
