A hybrid approach to beach erosion mitigation and amenity enhancement, St Francis Bay, South Africa

- Description: The St Francis Bay beach has experienced chronic erosion over the past three decades. This erosion can largely be attributed to the stabilisation of a large coastal dunefield which contributed +/- 80 percent of the sand supply to St Francis Bay. Stabilisation began in 1975 initially using plant cuttings and followed by the development of the Santareme holiday suburb resulting in complete stabilisation by 1985. Effects were felt from the late 1970‟s and since then the beach has retreated at between 0.5 - 3 m.yr-1. Erosion has encroached on beachfront properties since the early 1990‟s, leading to the placement of 3-4 m high unsightly rock revetments along much of the beach. Where properly maintained these structures have proved successful in protecting the properties behind, however exacerbated erosion of areas in front and adjacent to these structures is evident. Currently no dry beach is present at high tide for most of the year, leading to a significant reduction in beach amenity value. Several technical studies to investigate remediation of this beach erosion problem have been conducted since the early 1990‟s. This study includes investigations into the processes and dynamics of the existing environment and evaluation of the effectiveness and impacts of several elements of a hybrid approach to coastal protection and amenity enhancement for St Francis Bay beach. This proposal incorporated: Multi-Purpose Reefs (MPR‟s) offshore, for coastal protection and amenity enhancement in terms of surfing; beach nourishment with sand from the Kromme Estuary and dune rehabilitation with appropriate native sand binding species. Extensive fieldwork and data collection were conducted, this included: a series of bathymetric surveys; diving surveys and a helicopter flight; sediment sampling; beach profiling and deployment of a wave/current meter. Analysis of these data provided a greater understanding of the existing environment and dynamics of St Francis Bay and provided reliable inputs for numerical modelling. Numerical and physical modelling was conducted to assess the existing processes and conduct MPR design testing. In addition calibrated hydrodynamic modelling of the Kromme Estuary was conducted in order to assess the impacts of sand extraction from the large sand banks within the mouth of the Kromme Estuary for use as beach nourishment. Comparison of bathymetric survey data collected by the author in 2005/06 with survey data collected by the South African Navy Hydrographic Office (SANHO) in 1952 suggest a major loss of sand from the bay, with a volume difference of some 8.8 X 106 m3 calculated. Greater losses were measured between 10-15 m water depths, with shallow areas of +/- 5 m water depth, remaining more stable. This can be attributed to the presence of shallow reef and rocky substrate through much of the bay at this depth range. Monthly RTK GPS survey data from September 2006 to September 2007 indicates a total loss of 40 000 m3 over this period with the greatest losses measured along the northern part of the beach. The greatest losses were measured after large long period waves from a southerly to south-easterly direction occurred in conjunction with equinox tides in mid March 2007. Sediment sampling at over 100 locations within the bay indicated a high percentage of reef (26 percent) and fairly consistent grain size in the fine to medium size class throughout much of the beach, bay and large sand bank within the estuary. While the majority of the South African Coast is exposed to the predominant south westerly winds and waves, St Francis Bay‟s orientation means that waves from a south easterly to easterly direction dominate. The results of the detailed numerical modelling of the hydrodynamics agree with previous calculations and modelling results which concluded that strong unidirectional longshore currents occur along the headland due to the oblique angle of wave incidence and the close to parallel angle of wave incidence along the beach leads to weak longshore currents of variable direction. Erosion along St Francis Bay beach is a result of cross-shore erosion due to large waves from a southerly to easterly direction. Detached breakwaters are the most effective form of coastal protection in these environments and MPR‟s offer additional benefits over traditional breakwater structures. Results of empirical calculations and numerical modelling indicate that the MPR‟s will provide effective coastal protection through the processes of wave dissipation, wave rotation, salient formation and alteration of nearshore circulation. Physical modelling results allowed the MPR design to be assessed and refined in terms of surfing amenity enhancement and construction constraints. In addition numerical modelling results indicate that impacts due to the extraction of up to 600 000 m3 of sand from the lower Kromme Estuary result in highly localised velocity reduction, mainly limited to the extraction areas. The calculated rate of sediment influx into the lower Kromme Estuary indicates that limited extraction, in the order of 20 000 – 40 000 m3 per year, should be sustainable in the long term. Sedimentation of the lower estuary over recent years has had negative recreational and ecological impacts, through reduced navigability and water exchange respectively. Therefore both the estuary and beach systems prove to benefit from this approach. Although not investigated in detail as part of this study, evidence from numerous projects worldwide indicates that foredunes help to trap wind-blown sand on the beach and form a buffer to storm erosion, therefore dune rehabilitation with native sand-binding plant species was recommended as the third element of the proposed remediation of St Francis Bay beach.

- Description: This study focuses on the development of InAs/GaInSb strained-layer superlattice structures by metal organic chemical vapour deposition (MOCVD), and deals with two aspects of the development of InAs/GaInSb SLS’s by MOCVD viz. the deposition of nano-scale (~100 Å) GaInSb layers, and the electrical characterization of unstrained InAs. The first part of this work aims to study the MOCVD growth of GaInSb layers in terms of deposition rate and indium incorporation on the nano-scale. This task is approached by first optimizing the growth of relatively thick (~2 μm) epitaxial films, and then assuming similar growth parameters during nano-scale deposition. The GaInSb layers were grown as part of GaInSb/GaSb quantum well (QW) structures. By using this approach, the GaInSb QW’s (~100 Å) could be characterized with the use of photoluminescence spectroscopy, which, when used in conjunction with transmission electron microscopy and/or X-ray diffractomery, proves useful in the analysis of such small scale deposition. It is shown that the growth rate of GaInSb on the nano-scale approaches the nominal growth rates determined from thick (~2 μm) GaInSb calibration layers. The In incorporation efficiency in nano-layers, however, was markedly lower than what was predicted by the GaInSb calibration layers. This reduction in indium incorporation could be the result of the effects of strain on In incorporation. The choice of substrate orientation for QW deposition was also studied. QW structures were grown simultaneously on both (100) and 2°off (100) GaSb(Te) substrates, and it is shown that growth on non-vicinal substrates is more conducive to the deposition of high quality QW structures. The second part of this study focuses on the electrical characterization of unstrained InAs. It is long known that conventional Hall measurements cannot be used to accurately characterize InAs epitaxial layers, as a result of parallel conduction resulting from surface and/or interface effects. This study looks at extracting the surface and bulk electrical properties of n-type InAs thin films directly from variable magnetic field Hall measurements. For p-type InAs, the situation is complicated by the relatively large electron to hole mobility ratio of InAs which tends to conceal the p-type nature of InAs thin films from Hall measurements. Here, this effect is illustrated by way of theoretical simulation of Hall data.

The initial work included the development of a novel synthetic method for 1R,2R-diaminocyclohexaneoxalato-platinum(II) (oxaliplatin), by using an essentially non-aqueous solvent medium and direct ligand exchange at elevated temperatures. This was done by a study of the kinetics of the reaction in a variety of conditions; such as relative reagent concentrations and ratios as well as solvent mixtures. An effective method was developed which could be applied industrially. An international patent was taken out on this method. Various amine complexes of platinum(II) were synthesized using chloro, bromo and oxalato groups as leaving groups. The non-leaving groups were selected having certain specific characteristics in mind. Novel mononitroplatinum(IV) complexes were synthesized, mostly with oxalato leaving groups. One of these in particular, had excellent anticancer behaviour. Another trichloromononitro complex was also synthesized with very good anticancer properties. Two international patents were filed for the latter two compounds. As far as possible, all compounds were studied by spectrometric, chromatographic and thermal methods. They were also tested against 3 cancer cell lines namely cervical (Hela), Colon (HT29) and Breast (MCF7) cancer cells.

Alternative products in the inhibition of te plant pathogen scleroyinia sclerotiorum on potato production

- Description: White mold caused by Sclerotinia sclerotiorum attacks a wide host range of broad-leafed plants which includes potatoes. Current control is limited to the use of chemicals, but biological control has emerged as an eco-friendly alternative. For the study, 19 bacterial strains and 18 fungal strains obtained from soil samples taken from a potato field naturally infested with the pathogen S. sclerotiorum were tested for the effects on S. sclerotiorum mycelium growth and sclerotia viability in vitro. A total of eight bacterial strains and six fungal strains proved to be effective in the inhibition of S. sclerotiorum mycelium growth on PDA plate using a dual culture technique. These antagonistic microbes were six Bacillus subtilis, Bacillus pumilis, Bacillus marisflavi, Fusarium solani, Fusarium equiseti, Fusarium chlamydosporum, Aspergillus niger, Aspergillus insuetus and Penicillium citrinum. There was a significant difference (p<0.05) in the interaction between S. sclerotiorum mycelium growth and the time after inoculation with a S. sclerotiorum mycelium plug for both bacterial and fungal soil isolates. An additional sclerotia viability test was carried out using four of the eight antagonistic bacteria, and six of the antagonistic fungi. The results showed that two Bacillus species, namely B. pumilis and B. marisflavi can effectively reduce sclerotia viability. The other two bacteria (both B. subtilis), recorded low percentage reduction in sclerotia viability. As for the six antagonistic fungi, the viability test proved to be less effective in determining sclerotia viability. However, the result of this study supports the use of bio-control agents, not only because they are environmentally friendly, but because they are also an effective way of controlling the plant pathogen, S. sclerotiorum.

Towards the bioremediation of the hypertrophic Swartkops Solar Salt-works

- Description: This thesis presents the results of three studies aimed at improving brine-quality at the Swartkops solar salt-works (Swartkops Sea Salt [Pty] Ltd) on the outskirts of Port Elizabeth, South Africa. This is a highly eutrophic salt-works, the management of which has become increasingly difficult in recent years. The fundamental problem is how best to operate the system at maximum capacity while limiting nutrient inputs from the nutrient-rich microtidal Swartkops Estuary. In the first study, brine-quality at several sites along the axis of the Swartkops Estuary, and the extent to which it is affected by a variety of factors, is compared. Sites were sampled on micro- and macrotidal time scales, and were selected by the management of the salt-works as possible locations for a new pump-house (for extracting brine from the estuary) for their salt-work operations at Swartkops and Missionvale. The study showed that there are incremental benefits to be had from moving the site of extraction downstream from its present position to a site closer to the mouth of the estuary, where the concentration of nutrients usually is lower and where salinity usually is higher. There is little to be gained from moving the site of extraction laterally, to the mouth of the Inlet from which brine currently is extracted, so that brine is extracted directly from the estuary itself. A set of models relating the concentrations of NH+ 4 , NO{u100000}3 , and PO34{u100000} to salinity is proposed. These take into account the influences of site and season and may be used to estimate the concentration of these nutrients from a measurement of salinity. The model for PO34{u100000} shows that it would be more damaging to the salt-works’ operations to pump “low”-salinity brine during the early months of summer than during autumn. Evidence is also presented to show that Wylde Bridge has no influence on nutrient concentrations in the estuary, with tidal flushing generally passing beyond the Wylde-Bridge break-point. The exceptionally heavy flooding of the estuary that occurred in September 2002 may, however, have biased this conclusion, because of its scouring effect. The second study concentrated on monitoring the effect of (1) decreasing pond depth and (2) increasing pond salinity—two readily available management tools—on brine quality at the salt-works. Pond depth throughout the salt-works was decreased by 40 cm, and the salinity of Pond 5, a pond in the middle of the system, was increased to 175 S. Both measures were kept in place for the duration of the study (Nov. 2002–Aug. 2004). The pond-depth experiment did not have the expected result, there being no evidence of the increase in microalgal growth in the water column that was predicted based on previous research. There was, however, a significant increase in benthic chlorophyll-a, and there was a general improvement in the condition of the sedimentary system of the salt-works. There was also a substantial decrease in particulate organic matter in the water column, with clear evidence that the remaining fraction was closely associated with living forms of particulate matter rather than with detritus. The pond-salinity experiment proves that there is a flourishing, and resilient, population of brine shrimp (Artemia salina L.) at the salt-works. Restocking the salina, or stocking it with a different strain of brine shrimp, is therefore not necessary. The results of this study show that the brine shrimp population at the salt-works needs salinities of greater than about 65–70 S to survive. As a living force they almost certainly need a protective salinity that is greater than about 120–140 S, perhaps even as great as 160 S. Brine shrimp thrived in the high salinity milieu of the experimental pond for the duration of the study, but dwindled from three other ponds of the system once their salinities fell to below 90 S, eventually to disappear from them, apparently completely, once salinity fell to below 65 S. The third and final study concentrated on establishing whether the products released by decomposing barley straw could be used in a solar salt-works to control macroalgal blooms without detrimentally affecting the benthic-mat. Previous research has shown that these products are effective inhibitors of macroalgal growth and that they remain effective under saline conditions. The results presented here show that the same products, or products released under similar conditions of decomposition, adversely effect both the structure and the function of the mat. Consequently, their use in a solar salt-works cannot be recommended.

- Description: This thesis presents the results of three studies aimed at improving brine-quality at the Swartkops solar salt-works (Swartkops Sea Salt [Pty] Ltd) on the outskirts of Port Elizabeth, South Africa. This is a highly eutrophic salt-works, the management of which has become increasingly difficult in recent years. The fundamental problem is how best to operate the system at maximum capacity while limiting nutrient inputs from the nutrient-rich microtidal Swartkops Estuary. In the first study, brine-quality at several sites along the axis of the Swartkops Estuary, and the extent to which it is affected by a variety of factors, is compared. Sites were sampled on micro- and macrotidal time scales, and were selected by the management of the salt-works as possible locations for a new pump-house (for extracting brine from the estuary) for their salt-work operations at Swartkops and Missionvale. The study showed that there are incremental benefits to be had from moving the site of extraction downstream from its present position to a site closer to the mouth of the estuary, where the concentration of nutrients usually is lower and where salinity usually is higher. There is little to be gained from moving the site of extraction laterally, to the mouth of the Inlet from which brine currently is extracted, so that brine is extracted directly from the estuary itself. A set of models relating the concentrations of NH+ 4 , NO{u100000}3 , and PO34{u100000} to salinity is proposed. These take into account the influences of site and season and may be used to estimate the concentration of these nutrients from a measurement of salinity. The model for PO34{u100000} shows that it would be more damaging to the salt-works’ operations to pump “low”-salinity brine during the early months of summer than during autumn. Evidence is also presented to show that Wylde Bridge has no influence on nutrient concentrations in the estuary, with tidal flushing generally passing beyond the Wylde-Bridge break-point. The exceptionally heavy flooding of the estuary that occurred in September 2002 may, however, have biased this conclusion, because of its scouring effect. The second study concentrated on monitoring the effect of (1) decreasing pond depth and (2) increasing pond salinity—two readily available management tools—on brine quality at the salt-works. Pond depth throughout the salt-works was decreased by 40 cm, and the salinity of Pond 5, a pond in the middle of the system, was increased to 175 S. Both measures were kept in place for the duration of the study (Nov. 2002–Aug. 2004). The pond-depth experiment did not have the expected result, there being no evidence of the increase in microalgal growth in the water column that was predicted based on previous research. Changes in the zonation patterns within and between beaches were also investigated. The highest concentration of taxa and individuals was found near the low-water mark with a gradual reduction of both towards the high-water mark. Some of the sandy beach community variables (abundance, density and species richness) showed significant difference between the sampling sessions but no significant differences were seen between the two beaches. The presence of three zones was noted using MDS plots and Cluster diagrams. However, using a SIMPER analyses, four zones with the possibility of a fifth was noted.

Sputum specimens (102) were assessed by PCR employing primers directed at the following genes: P. jiroveci: mitochondrial large subunit ribosomal RNA (mtLSUrRNA), dihydropteroate synthase (DHPS) and dihydrofolate reductase (DHFR), and for M. pneumoniae: 16S rRNA and P1 adhesin. Positive P. jiroveci samples were genotyped by PCR-SSCP (single-strand conformation polymorphism) targeting the: internal transcribed spacer region (ITS), intron of the nuclear 26S rRNA gene (26S), variable region of the mitochondrial 26S rRNA gene (mt26S) and β-tubulin gene (β-tub). Multi-drug resistant (MDR-TB) cultures grown in the presence and absence of four antibiotics (rifampicin, isoniazid, ethambutol and ofloxacin) were heat killed, stained with SYTO16 and Propidium Iodide and analysed using flow cytometry. Rifampicin resistance gene mutations were screened by PCR and DNA sequencing. Details of patient’s gender, age, HIV and M. tuberculosis status were provided by the hospitals. Women were seen to be at high risk for community-acquired P. jiroveci colonisation. Overall, prevalence of P. jiroveci was 55.1 percent (54/102 patients). P. jiroveci was mainly associated with HIV (25/102 P. jiroveci positive patients for which clinical data was available) and co-colonisation with M. tuberculosis was observed in 11 cases. Sequence analysis of DHPS and DHFR products found no resistance associated mutations. M. pneumoniae was detected in one patient. Four simple SSCP patterns were identified and there were no co-infections with other P. jiroveci strains. Nine M. tuberculosis samples [8 MDR-TB isolates (NHLS) and M. tuberculosis ATCC® 27294TM] were tested. There was a 53 percent (19 out of 36 tests) agreement of flow cytometry with the BACTEC MGIT 960. Mutations (at two specific codons, namely 516 and 531) in the rifampicin resistance-determining region (RRDR) of the rpoB gene were observed in eight M. tuberculosis isolates. Evaluation of methods for genotyping and drug susceptibility testing of PcP and TB are imperative for epidemiology and drug resistance studies, and impact on treatment protocols.

The use of environmentally benign solvents is amongst the areas of green chemistry that have received the most attention. In this context, imidazolium ionic liquids have been widely reported to offer high product yields, fast reaction rates, excellent selectivity and generally mild working conditions, when used as reaction media. In addition, concerns about costs of solvents and the long-term environmental impact that can potentially result when solvents are discarded after their use have led to focused investigations into solvent-free procedures, as reported in recent literature. We have set out to explore the extent to which these advantages could be realized within our research. Non-volatile, non-flammable imidazolium ionic liquids [bmim][I], [bmim][BF4] and [bmim][PF6] were used as green solvents in ferrocene chemistry. Ferrocenoate esters were synthesised efficiently by the respective DCC/DMAP-promoted reactions of ferrocenecarboxylic acid and substituted benzoic acids or, alternatively, the DMAP-promoted reactions of ferrocenoyl fluoride with a range of substituted phenols in [bmim][BF4] and [bmim][PF6]. High yields and short reaction times were achieved. In addition, the ionic liquid was reused several times without a reduction in product yields. Under solvent-free conditions, DCC/DMAP-promoted reactions provided high yields within 3 min of reaction. The possible rearrangement of one of the intermediates in these reactions was modelled theoretically using density function theory (DFT) at the B3LYP/6-31G* level of approximation. Catalyst-free esterification was achieved by the application of microwave radiation to the reaction of ferrocenoyl fluoride and a range of substituted phenols. All the reactions were complete after 1 min of irradiation and products were isolated in high yield. DPAT, HfCl4, Sc(OTf)3 and Al(OTf)3 were screened as catalysts for esterification in [bmim][BF4] and under solvent-free conditions at various temperatures. All attempts at esterification of ferrocenecarboxylic acid with alcohols and phenols were unsuccessful. The Suzuki cross-coupling reaction was carried out in [bmim][BF4]. The isolated yields are, however, poor and suffer from poor reproducibility with different batches of [bmim][BF4] used.

The best of the tested methods were used on actual PMD data. The effect of using polarization scramblers was also tested.

- Description: River flow is important in controlling phytoplankton distribution in estuaries. Data on the effect of river inflow on phytoplankton distribution patterns in temporarily open/closed estuaries is lacking. This study investigated the influence of river inflow on size-fractionated phytoplankton biomass (Chl a), community composition and environmental parameters measured monthly over three years in two temporarily open/closed estuaries in the Eastern Cape, South Africa. A once-off primary production study over an annual cycle was completed in the Van Stadens and Maitland estuaries. The study monitored physical, chemical and biological characteristics in both estuaries to examine the effects of changes in environmental factors and river inflow. Daily sampling of physico-chemical and biological variables from river to sea was carried out in the Van Stadens to investigate short-time scale effects of changes in environmental factors and river inflow on the phytoplankton biomass. Five and three stations in the main channel of the Van Stadens and Maitland estuaries respectively were sampled at 0.5 m below the water surface and 0.5 m above the sediment surface for biological and chemical variables and at the surface, 0.25 m and every 0.5 m thereafter for physical parameters. Five stations adjacent to the main channel along the estuary were monitored for groundwater macronutrient concentrations and five additional sites located within the upper catchment of the Van Stadens River were sampled on a quarterly basis over two years. Both estuaries were characterised by distinct hydrological conditions, an overwash, an open, a closed and a semi-closed mouth phase. Flooding in the Maitland and Van Stadens estuaries in 2001 and 2002 caused sediment scour, altered channel morphology and brought about breaching of the mouth. Flood driven mouth-breaching events occurred three and four times in each of the estuaries during the study. The mouth stayed open 20 – 25 percent and was closed 60 – 65 percent of the time. In the Van Stadens the closed overwash mouth condition occurred approximately 10 – 20 percent of the time while in the Maitland it occurred less with the semi-closed mouth condition occurring 10 – 20 percent of the time. Incidents related to mouth opening not associated with strong river floods occurred approximately 10 – 15 percent of the time, although in the Maitland a semi-closed mouth state persisted more frequently than in the Van Stadens Estuary. During flooding events salinity dropped to low levels (< 5 psu) but soon recovered to brackish conditions when river flow was reduced and marine water penetrated deep upstream. Reduction in river flow combined with marine sediment deposition resulted in the closure of the mouth. During closed mouth conditions strong onshore storm surges and spring high tides introduced marine water through overwash that kept salinity high. In both estuaries salinity showed a negative correlation with rainfall (R2 = 0.12), indicative of the strong influence of marine overwash that kept salinity high thus masking the influence of freshwater. High rainfall in the Van Stadens Estuary caused high levels of turbidity that reduced light penetration at depth. Light attenuation was positively correlated with the high rainfall (R 2 = 0.26) suggesting that increased turbidity was linked to rainfall induced discharge. In contrast, in the Maitland Estuary light attenuation did not show any correlation with increased rainfall possibly because of the reduced water depth and increased euphotic zone following the floods in 2002. High river inflow introduced macronutrients in both estuaries such that dissolved inorganic phosphates (DIP) and dissolved inorganic nitrogen (DIN) concentrations in the Van Stadens Estuary were strongly correlated with rainfall (R2 = 0.78 and 0.57 respectively). In the Maitland Estuary DIP and DIN concentrations remained significantly higher (p < 0.05) compared to that in the Van Stadens suggesting that the Maitland catchment contributed greater nutrient input into the estuary and may be associated with farming activities. Phytoplankton chlorophyll a (Chl a) ranged from 0.8 – 13.9 μg L-1 in the Van Stadens and in the Maitland Estuary from 5.3 – 138 μg L-1 during the 3-year study. During the open mouth condition Chl a biomass and primary production ranged from 5.4 – 52.9 μg Chl a L-1 and 1.2 – 11.7 mg C m-2 d-1 in the Maitland and in the Van Stadens from 1.6 – 9.8 μg Chl a L-1 and 1.2 - 14 mg C m-2 d-1 respectively. Maximum annual primary production in the Maitland and Van Stadens estuaries was 8.8 and 5.1 g C m-2 y-1 respectively. When the mouth was open in the Van Stadens Estuary the microphytoplankton (> 20 μm) accounted for > 65 percent of the Chl a, whereas during closed mouth conditions they accounted for about 55 percent of the Chl a biomass. Chlorophytes became the dominant taxon in the dry summer months but were replaced by cryptophytes and dinoflagellates during the wet season. When nutrient concentrations were low during low flow conditions in the Van Stadens Estuary mixotrophic microphytoplankton became an important fraction of the water column together with phototrophic dinoflagellates and cryptophytes. High river inflow initially reduced Chl a biomass followed by a recovery period of a couple of days compared to a 8 – 10 week recovery period in studies monitored over seasonal and annual temporal scales. The responses may be dissimilar but help to illustrate that there are similar response patterns to environmental forcing necessary to support phytoplankton biomass at different temporal scales. This study has demonstrated that flooding events caused by strong river flow cause breaching of the mouth, a reduction in salinity and marked nutrient input. Although the causes of flooding can be similar in both estuaries the resultant effects are varied and can alter the ability of the estuary to retain water. This study was able to demonstrate that the supply of macronutrients from the catchment was strongly correlated with rainfall (R2 = 0.67) and that phytoplankton growth mainly depended on an allochthonous source of macronutrients although internal supplies could be critical at times in controlling microalgal biomass.

- Description: River flow is important in controlling phytoplankton distribution in estuaries. Data on the effect of river inflow on phytoplankton distribution patterns in temporarily open/closed estuaries is lacking. This study investigated the influence of river inflow on size-fractionated phytoplankton biomass (Chl a), community composition and environmental parameters measured monthly over three years in two temporarily open/closed estuaries in the Eastern Cape, South Africa. A once-off primary production study over an annual cycle was completed in the Van Stadens and Maitland estuaries. The study monitored physical, chemical and biological characteristics in both estuaries to examine the effects of changes in environmental factors and river inflow. Daily sampling of physico-chemical and biological variables from river to sea was carried out in the Van Stadens to investigate short-time scale effects of changes in environmental factors and river inflow on the phytoplankton biomass. Five and three stations in the main channel of the Van Stadens and Maitland estuaries respectively were sampled at 0.5 m below the water surface and 0.5 m above the sediment surface for biological and chemical variables and at the surface, 0.25 m and every 0.5 m thereafter for physical parameters. Five stations adjacent to the main channel along the estuary were monitored for groundwater macronutrient concentrations and five additional sites located within the upper catchment of the Van Stadens River were sampled on a quarterly basis over two years. Both estuaries were characterised by distinct hydrological conditions, an overwash, an open, a closed and a semi-closed mouth phase. Flooding in the Maitland and Van Stadens estuaries in 2001 and 2002 caused sediment scour, altered channel morphology and brought about breaching of the mouth. Flood driven mouth-breaching events occurred three and four times in each of the estuaries during the study. The mouth stayed open 20 – 25 percent and was closed 60 – 65 percent of the time. In the Van Stadens the closed overwash mouth condition occurred approximately 10 – 20 percent of the time while in the Maitland it occurred less with the semi-closed mouth condition occurring 10 – 20 percent of the time. Incidents related to mouth opening not associated with strong river floods occurred approximately 10 – 15 percent of the time, although in the Maitland a semi-closed mouth state persisted more frequently than in the Van Stadens Estuary. During flooding events salinity dropped to low levels (< 5 psu) but soon recovered to brackish conditions when river flow was reduced and marine water penetrated deep upstream. Reduction in river flow combined with marine sediment deposition resulted in the closure of the mouth. During closed mouth conditions strong onshore storm surges and spring high tides introduced marine water through overwash that kept salinity high. In both estuaries salinity showed a negative correlation with rainfall (R2 = 0.12), indicative of the strong influence of marine overwash that kept salinity high thus masking the influence of freshwater. High rainfall in the Van Stadens Estuary caused high levels of turbidity that reduced light penetration at depth. Light attenuation was positively correlated with the high rainfall (R 2 = 0.26) suggesting that increased turbidity was linked to rainfall induced discharge. In contrast, in the Maitland Estuary light attenuation did not show any correlation with increased rainfall possibly because of the reduced water depth and increased euphotic zone following the floods in 2002. High river inflow introduced macronutrients in both estuaries such that dissolved inorganic phosphates (DIP) and dissolved inorganic nitrogen (DIN) concentrations in the Van Stadens Estuary were strongly correlated with rainfall (R2 = 0.78 and 0.57 respectively). The techniques showed good agreement in the measured PMD value, both in the laboratory and field measurements. In particular very good agreement was found between the JME average DGD and the extrema counting analysis PMD value. The GINTY and FA Fourier analysis method also gave very similar PMD values. It was found that the fibre birefringence and the mode coupling manifest in different ways on the intensity spectrum. By using the FA ratio method, the length regimes of the different fibre types were determined. Three characteristics of the FA technique were investigated, namely: wavelength window variation, sampling and input SOP scrambling. It was found that the wavelength window and the PMD are inversely proportional. Correct sampling plays a significant role in determining the correct measured PMD value. Lastly an average PMD value over the PMD values for different input SOPs serves as a better representation of the true PMD value. An additional study showed that the FA technique and a developed Poincaré sphere analysis method agree very well regarding the PMD value.

- Description: The work presented in this thesis is concerned with the evaluation of potential synthetic routes for the diacylhydrazine group of compounds, and particularly, unsymmetrical diacylhydrazines. Diacylhydrazines form the basis for a relatively new group of insecticides that have molt accelerating properties, and which are considered to offer substantial advantages over other insecticides used for the control of certain insects. The overall objective for this study is to evaluate different potential synthetic routes for a model diacylhydrazine with the view to define potentially scaleable routes. The compound selected for this study was the unsymmetrical diacylhydrazine, N-[N-(tertbutyl) phenylcarbonylamino](4-methylphenyl)-carboxamide since it offers the same range of challenges that would be expected for the synthesis of other similar unsymmetrical diacylhydrazines. Thus, the synthesis of unsymmetrical diacylhydrazines require two reaction steps: The first step is the synthesis of the intermediate mono-acylhydrazine, while the second step is the synthesis of the desired diacylhydrazine from the intermediate mono-acylhydrazine. The most important factor in the two-step reaction sequence is to obtain a high degree of selectivity for the desired mono-acylhydrazine isomer. Acylation of t-butylhydrazine using 4-methylbenzoyl chloride can produce the desired product N-[(tert-butyl)amino](4-methylphenyl)carboxamide in yields above 90 percent, but this reaction produces a variety of by-products, including the “wrong” isomer (N-amino-N- (tert-butyl)(4-methylphenyl)carboximide). Unexpected byproducts for this particular acylation reaction, not previously reported in the literature have also been identified during this investigation. This includes a de-butylated diacylhydrazine, (4-methylphenyl)- N-[(4-methylphenyl)carbonylamino] carboxamide. Although the reaction between tert-butylhydrazine and 4-methylbenzaldehyde is very selective giving near quantitative yields of the desired hydrazone, the subsequent conversion of the hydrazone into the desired mono-acylhydrazine is problematic. The most promising route appears via bromination to form the hydrazidic bromide, followed by hydrolysis of the hydrazidic bromide. Yields for the bromination reaction during this investigation were somewhat higher than that reported previously in literature. Hydrolysis of the hydrazidic bromide, apparently also results in the hydrolysis of the reaction product to give an ester of the free acid (when an ester solvent is used). On the optical characterization of photovoltaic devices

- Description: Semiconductor materials used for making photovoltaic (PV) cells have defects and impurities due to constraints of keeping production costs low. Electrically active defects that are distributed over the bulk material lead to reduced overall performance of a photovoltaic (PV) cell. In this study an adaptable Light Beam Induced Current (LBIC) system was designed in order to characterize the local performance of PV cells. The system uses a laser source and objective lens mounted vertically above the sample on the X-Y stage. Two current pre-amplifiers are used for converting the photo-generated current from the PV cell and the signal from the reflection detector cell into a voltage that can be measured by the data acquisition board. Two configurations for measuring the photo-current maps of either bare cells or encapsulated PV mini-modules can be used. To add flexibility it was envisaged that the system would be built in such a way that it allows easy integration of carrier lifetime mapping capabilities while keeping costs to a minimum. The carrier lifetime measurement technique integrated into the LBIC system is based on the optical Open-Circuit Voltage-Decay (OCVD) method. In a single-crystalline silicon PV cell that was tested, photo-current and opencircuit voltage LBIC scans revealed shunting behaviour due to scratch marks on the front surface. The marks are believed to have been caused by poor handling during manufacturing process. Reduced photo-current due to edge shunting was observed towards the edges of the PV cell. In another sample of single crystalline silicon cell an edge shunt resulted in a 30 percent drop in photo-current measured. LBIC measurements performed on multi-crystalline cells revealed nonuniformities such as enhanced photo-current on one side of grain boundaries. These asymmetric enhancements of local photo-current are attributed to the incline of the grain boundary into the bulk of the material. LBIC results obtained from mini modules showed a high degree of mismatching from cell to cell. It is well known that mismatching can degrade the performance of a PV module with series connected cells. The LBIC measurements presented also illustrated the negative effects of delamination on the photo-current of PV modules. The LBIC measurements performed on mini PV modules highlight some of the benefits of using an LBIC technique as a tool for investigating PV cell’s local photo-current response. The effective lifetime maps obtained highlight the importance of optical OCVD method as a tool that can be used in conjunction with conventional LBIC technique. Effective minority carrier lifetimes around 40 ms were measured on a single crystalline silicon cell of 2.5 x 2.5 cm2. Local features such as cracks and lifetime degrading defects were revealed by LBIC maps and were also confirmed on effective lifetime maps. The results presented demonstrate the importance of using effective carrier lifetime maps to complement photocurrent maps of PV cells and identification of areas where defects are located.

- Description: In previous work done in our laboratories, a method was discovered to produce phenolic mono-ethers from 4-hydroxyacetophenone and other 4-hydroxyketones by treating with ammonium peroxy-disulfate in an alcohol as a reaction solvent and in the presence of concentrated sulphuric acid or other strong protonic acids. Since this method of producing 4-alkoxyphenol ethers provides a very convenient way to modify hydroquinone and substituted hydroquinones to produce a variety of phenol mono-ethers, it was of interest to study the general scope of this reaction, including a more detailed investigation of the reaction mechanism. In previous studies, it was suggested that interaction between the aromatic pi-system of hydroquinone and the cyclohexa-2,5-diene structure of benzoquinone plays a significant role during the reaction. It was therefore of interest to investigate whether other compounds that are also capable of forming the cyclohexa-2,5-diene structure, will interact in a manner analogous to the hydroquinone/benzoquinone couple. Two specific compounds were selected for this purpose, namely 4-nitrosophenol and 4- (diphenylmethylene)cyclohexa-2,5-dien-1-one . The scope of etherification reactions of hydroquinone-benzoquinone or hydroquinone/benzoquinone like substrates such as 4-nitrosophenol and 4- (diphenylmethylene)cyclohexa-2,5-dien-1-one in the presence of acid catalyst and alcohols was investigated. These studies showed that hydroquinone, 4- nitrosophenol and 4-(diphenylmethylene)cyclohexa-2,5-dien-1-one successfully affords the phenolic ethers in good to excellent yield. For example, quantitative yield of 4-methoxyphenol could be obtained from a 1:1 mixture of hydroquinone and benzoquinone at the reflux temperature of methanol. In order to study the reaction mechanism, the cross-over reaction between tertbutylhydroquinone and benzoquinone (or hydroquinone and tertbutylbenzoquinone) was studied in detail. The results of these cross-over reactions were used to propose a mechanistic pathway that could explain the requirement for pi-interaction between the hydroquinone and benzoquinone molecules, the role of the acid catalyst, as well as the relative rates of hydroquinone and benzoquinone consumption during these reactions. The mechanism was also capable of explaining all the reaction products observed during these reactions. The work was then extended to reactions of 4-nitrosophenol and 4- (diphenylmethylene)cyclohexa-2,5-dien-1-one with methanol in the presence of either hydroquinone or benzoquinone. The results of these investigations strongly suggest the presence of similar interactions between these molecules that also influence the outcome of the reactions. The exploitation of pi-interactions between two molecules of these types investigated during this work opens an interesting field of chemistry. Clearly, the level of understanding developed during this work is only beginning to address this interesting field of chemistry and much work will need to be done to gain a fuller understanding of the chemistry involved as well as the potential synthetic value of these interactions.

- Description: This study presents the results of a study on introduced giraffe diet and feeding effects within the Eastern Cape Province of South Africa. Five sites were selected over a moisture gradient from the mesic east to the arid west of the Eastern Cape to describe giraffe diet and provide baseline data for feeding effect experiments. At each site faecal samples were collected seasonally and diet composition determined by microhistological analysis. Plant species availability was measured using the point intersect method in order to compare plant availability to plant consumption to determine diet preferences. Giraffe diet across the sites showed a unimodal response to rainfall by utilising fewer species in drier areas where plant availability is low and increasing the number of species consumed as the rainfall increases. In higher rainfall areas, giraffe reduced the number of plant species fed upon because of the high availability of important species that reduces the need for a varied diet. Across all sites, plant availability was dominated by woody plant species (more than 90 percent at each site). A total of 57 plants species were recorded as being eaten across the five sites. Several PDI (Principle Diet Items) plant species (Acacia karroo, Schotia afra, Pappea capensis and Euclea undulata) were eaten across sites. Over all the sites, significantly preferred species were Asparagus striatus, Schotia latifolia, Asparagus suaveolens, Commelina benghalensis, Viscum rotundifolium, and Acacia cyclops. Acacia karroo, Schotia afra, Pappea capensis, Rhus crenata, A. tetracantha, and Grewia robusta were utilised in proportion to their availability. Euclea undulata, Rhus longispina and Putterlickia pyracantha were avoided. Feeding effects were tested by erecting exclosures around trees, covering one half of each tree. Ten trees were selected for these manipulative experiments which ran from September 2003 to February 2005. The number of branches, number of leaves, branch orders, branch diameter, branch length, leaf length and leaf width were measured for ten samples per side (enclosed vs. exposed) and analysed using a Wilcoxon matched pairs test and tested for effect size using Cohen’s d. Although no significant differences were detected between the sides measured, the effect size indicated differences in all measurements between sides ranging from low to moderate. Leaf length was greater on the outside of exclosures and showed a moderate difference between the sides in terms of the d-value with the p-value (0.059) tending towards significance. Although leaf width was higher on the inside of exclosures, it only showed a moderate difference for d with no statistical significance. Leaf area was higher on the inside of exclosures and tended to significance (p = 0.059) and similarly the d value indicated moderate differences between the sides in terms of effect size. Similarly branch length was higher on the inside of exclosures although not significantly, the p-value (0.07) approached significance. The effect size for branch length also showed a moderate difference between the sides. The number of branches and the number of leaves showed no statistical differences between the sides although the p-value (0.059) for the number of branches, as well as number of leaves (p = 0.07) approached significance. For both these variables, effect size showed a moderate difference. Giraffe showed diversity in their diet selection across sites but mainly fed upon available species. It is concluded that giraffe impact on sites may be reduced if populations are properly managed however, high densities of giraffe could lead to similar vegetation impacts as observed in other areas where giraffe have been introduced and potentially change vegetation structure and plant community composition.

An adaptive user interface model for contact centres

- Description: Contact centres (CC), are the primary interaction point between a company and its customers and these are rapidly expanding in terms of both workforce and economic scope. An important challenge for today's CC solutions is to increase the speed at which CCAs retrieve information to answer customer queries. CCAs, however, differ in their ability to respond to these queries and do not interact with the computer user interface (UI) in the same way as they each have different capabilities, experience and expertise. Studies have provided empirical support that user performance can be increased when the computer UI characteristics match the user skill level. Adaptive user interfaces (AUIs) are the key to creating personalised systems. Their sole task is to provide an interface most suitable to users' needs whilst facilitating the users' varying skill levels. The aim of this research was to develop an AUI model for CCs to support and improve the expertise level of CCAs. A literature review of CCs, user expertise, AUIs and existing AUI models resulted in the proposal of an AUI model for CCs. The proposed AUI model was described in terms of its architecture, component-level and interface design. An AUI prototype was developed as a proof-of-concept of the proposed AUI model. A literature review on existing AUI evaluation approaches resulted in an evaluation strategy for the proposed AUI model. The AUI prototype was evaluated according to the evaluation strategy that was identified. User testing incorporating eye-tracking and a post-test questionnaire was used to determine the usefulness and usability of the AUI prototype. Significant results were found with regards to user satisfaction ratings, the learnability of the AUI prototype and its effectiveness. This dissertation makes an important contribution in the design of an AUI model that supports and improves the expertise level of CCAs. The model could be used to assist the development of CC applications incorporating AUIs. Future research is however needed to evaluate the effect of the proposed AUI model in a larger CC environment.

Development of platinum metal specific separating agents

- Description: In this dissertation, the aim was to develop a platinum specific resin to be utilized for the early removal of platinum from the industrial feed solution. Efforts were therefore directed towards the syntheses of silica based resins, with active centra, designed for platinum. The large chlorometallate ions present in the feed stream, were characterized in terms of physical parameters relevant to phase distribution, namely distortability (RD), charge density, softness (σ) etc. Matching cations for each of the types were investigated. In order to attempt the design of platinum specific resins, different structural amines were used to aminate the silicone precursor and to subsequently fix these onto the silica framework. Two different solvents namely alcohol and dmf were used in this process, resulting in two sets of resins, with different properties. The design was based on previous experience with these ions, with reference to their behaviour towards different types of cations. The platinum species, PtCl6 2- and PtCl4 2-, as well as the most important contaminants in the feed stream, were typified, bearing in mind size, charge, charge density and distortability. Different types of cationic centra, having differences in charge density, stereochemical crowding and extent of hydrophobicity, were synthesized and tested-both as solvent extractants (where possible) and silica based resins. The results indicated that, partly screened secondary ammonium cationic resin species, which could be regarded as “intermediate”, proved to be satisfactory both in their high percentage extraction for PtCl4 2- and rejection of contaminants like chlororhodates, chloroiridates(III) and FeCl4 -. It was however necessary, to work at a redox potential, where iridium(IV) in the form of IrCl6 2-, was absent. Various 2-aminoalkane resins were prepared, with variation in the length of alkane group and synthesised by the two different solvents. The latter resulted in two sets of resins with not only differing compactness, but also having significantly different properties with reference to platinum specificity, HCl effect and stripping potential. The 2-aminobutane and 2-aminoheptane resins in particular, proved to be very satisfactory platinum specific resins, both with respect to selectivity, platinum capacity and stripping potential. The various physical parameters could be applied to identify the chemical behaviour of anions and assist in the development of anion specificity for the relevant species.

- Description: In this dissertation, the aim was to develop a platinum specific resin to be utilized for the early removal of platinum from the industrial feed solution. Efforts were therefore directed towards the syntheses of silica based resins, with active centra, designed for platinum. The large chlorometallate ions present in the feed stream, were characterized in terms of physical parameters relevant to phase distribution, namely distortability (RD), charge density, softness (σ) etc. Matching cations for each of the types were investigated. In order to attempt the design of platinum specific resins, different structural amines were used to aminate the silicone precursor and to subsequently fix these onto the silica framework. Two different solvents namely alcohol and dmf were used in this process, resulting in two sets of resins, with different properties. The design was based on previous experience with these ions, with reference to their behaviour towards different types of cations. The platinum species, PtCl6 2- and PtCl4 2-, as well as the most important contaminants in the feed stream, were typified, bearing in mind size, charge, charge density and distortability. Different types of cationic centra, having differences in charge density, stereochemical crowding and extent of hydrophobicity, were synthesized and tested-both as solvent extractants (where possible) and silica based resins. The results indicated that, partly screened secondary ammonium cationic resin species, which could be regarded as “intermediate”, proved to be satisfactory both in their high percentage extraction for PtCl4 2- and rejection of contaminants like chlororhodates, chloroiridates(III) and FeCl4 -. It was however necessary, to work at a redox potential, where iridium(IV) in the form of IrCl6 2-, was absent. Various 2-aminoalkane resins were prepared, with variation in the length of alkane group and synthesised by the two different solvents. The latter resulted in two sets of resins with not only differing compactness, but also having significantly different properties with reference to platinum specificity, HCl effect and stripping potential. The 2-aminobutane and 2-aminoheptane resins in particular, proved to be very satisfactory platinum specific resins, both with respect to selectivity, platinum capacity and stripping potential. The various physical parameters could be applied to identify the chemical behaviour of anions and assist in the development of anion specificity for the relevant species.

The effect of different management practices on the feed intake and growth rate of ostrich chicks

- Description: Different management techniques were studied, at the Kromme Rhee Experimental Farm near Stellenbosch, in order to stimulate feed intake of ostrich chicks and to determine the effect on their growth rate. A first experiment was conducted to determine the effect of different dietary flavourants on the feed intake of ostrich chicks. Ostrich chicks (1-28 days of age), with no previous exposure to flavourants showed a strong preference for salty feed. Another experiment determined the effect of different colourants on the feed intake of ostrich chicks. Exposure of chicks to feed coloured artificially in different colours (green, red, blue and yellow) had no impact on the feed intake of ostrich chicks (1-28 days of age). Throughout the study the dry matter intake was the highest for the control diet, indicating that the provision of coloured feed to ostriches, as practised in this study, will not stimulate feed intake. The effect of feed processing and restriction of water availability for ostrich chicks was also examined. Extrusion reduced microbial counts in feed by 99.7 percent. Feed intake as well as average daily gain of chicks were lowered by both the pelleting and the extrusion process practised in this study. Processing did not have a positive influence on the production of ostrich chicks 6-15 weeks of age. Dry matter intake for chicks subjected to water restriction were the same as for birds receiving water ad libitum. Similarly growth rate as well as feed conversion ratio was not effected by restriction of water, as practised in this study. Another technique studied was to determine the effect of 24h artificial light on feed intake, growth and the behaviour of ostrich chicks (1-28 days of age). A Timeactivity budget revealed that chicks subjected to 24 hours of light spent more time eating (14.7 percent) than those subjected to normal day and night cycles (11.4 percent). In spite of this, no significant difference in feed intake, growth rate and feed conversion ratio was observed for ostrich chicks subjected to different light regimes. The effect of different dietary salt levels on the production of ostrich chicks (8-12 weeks of age) was also determined. The performance of chicks was the best on a diet with a salt inclusion of 1.4 percent.
