Equally, the density of physiologically stressed stems differed substantially in the three areas with higher in FC 20.5± 75.5 stems/ha compared to 18.7±82.9 stems/ha in TR and 3.8±11.8 stems/ha in LM. This results suggests implementation of protection measures such as the inclusion of the species in the national CITES list to regulated cross-border trade and strengthening of the law enforcement. Genetic diversity, structure and gene flow in the exiting populations of southern Mozambique revealed that despite fragmentation and overexploitation, this species maintains a relatively high level of genetic diversity supporting the existence of random mating. Two genetic groups were found corresponding to the northern (LM and TR) and southern (FC) areas. The study suggests that, if local extinctions occurred in Mozambique, the pepper-bark tree persisted in sufficient numbers to retain a large proportion of genetic diversity. During the present study in the Sundays Estuary, low oxygen waters were recorded in the middle reaches (Site S5) mostly during summer (four months of low DO conditions). In the Swartkops Estuary, low dissolved oxygen was recorded in the upper reaches during spring. The lowest dissolved oxygen concentration recorded was 0.5 mg/l and 2.4 mg/l in the bottom waters of the Sundays and Swartkops estuaries, respectively. Selected dominant species were only absent from areas where dissolved oxygen was < 1 mg/l and present in the adjacent sites (for example Site S4, and S3) where DO was higher mostly during January 2019. As such, the low dissolved oxygen concentrations recorded in the Swartkops Estuary did not have a noticeable impact on fish distribution, although the total abundance of species did show a slight decline when dissolved oxygen was < 3 mg/l. This study demonstrates the importance of concurrently examining estuarine and nearshore marine habitats in order to identify ecologically important habitats, which has important implications for the development of effective management strategies for coastal fish populations, particularly in the light of anthropogenic change. In addition, in order to identify nursery hotspots it is crucial to correctly identify all the species occupying these areas. Similarly, in the α-glucosidase inhibition assay, all the four derivatives exhibited a concentration dependent activity with TZDD3 showing the most activity. All the four derivatives exhibited ≥ 30% inhibition in the aldose reductase inhibition assay except TZDD1 at 10 μg/mL. TZDD4 exhibited a concentration dependent inhibition activity in the protein tyrosine phosphatase-1B inhibition assay. Interestingly, TZDD3 showed a decreasing inhibition activity as its concentration increased from 10 μg/mL through to 50 μg/mL. In the dipeptidyl peptidase–4 inhibition assay, TZDD2 and TZDD4 exhibited ≥ 20% inhibition activity across all the concentrations and in the acetylcholinesterase assay, TZDD1, 3 and 4 exhibited ≥ 25% across all the concentrations. Interestingly, in the matrix metalloproteinase-1 inhibition assay, some of these derivatives exhibited partial activation activity and partial inhibition with TZDD1 showing activation in concentrations 10 and 20 μg/mL and inhibition in concentrations 30, 40 and 50 μg/mL. TZDD4 showed activation in all the concentrations. In the β-amyloid aggregation assay, all the four derivatives showed inhibition activity ≥ 10% except TZDD1 at 50 μg/mL. Conclusions Diabetes mellitus and Alzheimer’s disease are a type of pathology of global concern, and several researchers worldwide have strived to search for novel therapeutic treatments and prevention for diabetes as well as Alzheimer’s disease. Recent studies provide a direct link v between diabetes mellitus and Alzheimer’s disease, and the need to find novel drugs that can mitigate these two is of increasing interest. In our search for antidiabetic and anti-Alzheimer’s activity, TZD derivatives (TZDD1, TZDD2, TZDD3 and TZDD4) exhibited good antioxidant activity, anti-hyperglycaemic activity and a relatively promising anti-Alzheimer’s activity. This was observed from the in vitro evaluation performed which included α – amylase, α – glucosidase, aldose reductase, PTP1B, DPP4, amyloid β aggregation, and AChE inhibition assays. Furthermore, docking of the derivatives against PPARγ predicted a similar molecular interaction to that of thiazolidinediones using Rosiglitazone as the standard drug. Furthermore, in silico ADME profiling predicted the derivatives to have moderate to good solubility in the GI (good GI bioavailability), and also exhibited excellent drug likeness. However, they are predicted not permeate the BBB. Degradation is dominant in the upper (T35B and T3C) and lower (T35K, T35L and T35M) parts of the catchments. The three criteria used by the stakeholders in the prioritisation process of the key EI were assigned 12 spatial attributes (the catchment characteristics about the study area in relation to the criteria) to indicate relevant information needed for selecting suitable restoration areas to enhance flow regulation. The AHP analysis results identified approximately 63% (17,703 ha) of wetlands, 88% (235,829 ha) of grasslands, 78% (13,608 ha) of abandoned cultivated fields and 93% (3,791 ha) of the riparian zones as suitable areas for restoration to mitigate drought impact through flow regulation. Also, the suitability results showed 63% (17,703 ha) of wetlands, 58% (2,203 ha) of riparian zones, 68% (11,745 ha) of abandoned cultivated fields and 46% (122,285 ha) of grasslands as suitable restoration areas for improving ecosystem services for community livelihoods. The AHP analysis identified more than 39-43% (of the degraded EI indicated by the Trends.Earth analysis) areas that are suitable for restoration, because key EI plays a significant role in flow regulation and people’s livelihoods, especially when they are managed, maintained, and restored to good health conditions. Therefore, the prioritized EI areas should be either maintained, managed, rehabilitated or restored. The major distinct causes of land degradation are woody encroachment in grasslands, invasion of alien plants on abandoned cultivated fields and soil erosion in the catchment. The most suitable EI areas recommended for restoration are those natural resources near local communities, which provide essential ecosystem services to sustain their livelihood. Therefore, degraded EI in the T35 catchments should be restored and maintained to improve livelihood and mitigate drought impacts. The study pointed out how the key selected ecological infrastructure can help mitigate the impacts of droughts and improve human livelihood.
